表面抗原调节慢性乙肝患者树突状细胞免疫功能的研究

【摘要】 目的研究表面抗原(HBsAg)对慢性乙型肝炎(CHB)患者外周血树突状细胞(DC)免疫功能的调节作用。方法采用体外细胞培养的方法培养30例CHB患者外周血DC,分为HBsAg组和无HBsAg组,培养7d后用流式细胞术检测各组DC的CD83及CD86的表达,MTT法检测DC对混合淋巴细胞的刺激作用,ELISA方法检测培养上清中IL-12及IL-6的含量。结果 HBsAg组CD83、CD86的表达量分别为(19±4)%和(49±9)%,无HBsAg组则分别为(12±4)%和(18±6)%(P均<0.01)。HBsAg组和无HBsAg组的DC对混合淋巴细胞的刺激指数分别为(20±10)%和(7±4)%(P<0.01)。HBsAg组的DC培养上清中IL-12、IL-6的含量分别为(111±68)ng/ml和(646±426)ng/ml,无HBsAg组分别为(38±12)ng/ml和(1843±235)ng/ml,差异均有统计学意义(P均<0.01)。结论 HBsAg可增强CHB患者外周血DC提呈抗原、激活T淋巴细胞的功能,对DC分泌细胞因子的功能有调节作用。

【Abstract】 Objective To investigate the regulative effect of HBsAg on the immune function of dendritic cells(DCs) in patients with chronic hepatitis B(CHB) in vitro.Methods Peripheral blood DCs in 30 patients with CHB were cultured in vitro.They were divided randomly into two groups,HBsAg group and control group.Expression of CD83 and CD86 on DCs was analyzed by flow cytometry.The capability of DCs stimulating T lymphocyte proliferation was evaluated by allogenetic mixed lymphocyte reaction.IL-12 and IL-6 levels in cell culture supernatant were detected by ELISA.Results The expression of CD83 and CD86 on DCs in HBsAg group was(19±4)%and (49±9)%,compared to(12±4)%and(18±6)%in control group(P<0.01),respectively.Stimulation index of DCs to T lymphocytes was(20±10)%in HBsAg group and(7±4)%in control group(P<0.01).The levels of supernatant IL-12 and IL-6 were(111±68) pg/ml and(646±426) pg/ml in HBsAg group,compared to(38±12) pg/ml and(1843±235) pg/ml in control group(P<0.01).Conclusion HBsAg can enhance the capabilities of antigen presentation,activate T lymphocytes of DCs in patients with CHB,and regulate the capability of secreting cytokines in DCs.
