【摘要】 目的:研究TRPV6在子宫内膜癌组织的表达及其与临床病理特征的关系。方法:选取58例子宫内膜癌石蜡切片,用免疫组化法测定TRPV6的分布及表达,分析其与临床病理之间的关系;另取31例正常子宫内膜及13例不典型增生内膜石蜡切片为对照。选取15例子宫内膜腺癌及11例正常子宫内膜新鲜组织标本,以实时定量PCR方法检测TRPV6的表达。结果:(1)TRPV6在子宫内膜癌组织的表达主要位于胞膜及胞浆内,阳性表达率77.6%,显著低于非绝经期子宫内膜组织(100%)及不典型增生子宫内膜组织(100%)(P<0.05),且前者表达强度显著低于后二者(P<0.01)。mRNA水平研究表明TRPV6在内膜癌组织中的表达也显著低于正常内膜组织(P<0.05);(2)绝经后子宫内膜癌患者TRPV6阳性表达率为71.8%,显著低于未绝经患者(89.5%);(3)在子宫内膜癌组织中,宫颈受累者TRPV6阳性率显著高于宫颈未受累者(89.3%vs 67.7%,P=0.039)。结论:TRPV6在子宫内膜癌组织的表达低于正常子宫内膜组织,TRPV6阳性表达者较多发生子宫内膜癌宫颈受侵,表明钙通道蛋白TRPV6与子宫内膜癌有一定的关系,为探讨子宫内膜癌发病分子机制提供了研究资料。更多还原

【Abstract】 Objective:The expression of transient receptor potential vanilloid 6(TRPV6)in endometrial cancer was detected and its relationship with clinicopathological features was analyzed.Methods:The expression of TRPV6 was detected in endometrial carcinoma,atypical endometrial hyperplasia and normal endometrium by immunohistochemistry and real-time quantitative PCR.Results:(1)TRPV6 expression was mainly located in the membrane and cytoplasm of endometrium.The positive rate of TRPV6 expression was 77.6% in endometrial cancer.It was significantly lower than that in premenopausal normal endometrium(100%)and atypical endometrial hyperplasia(100%)(P<0.05).Real-time quantitative PCR results also showed that the expression of TRPV6 was lower in endometrial cancer tissues than that in normal endometrial tissues in mRNA level(P<0.05).(2)The expression of TRPV6 was lower in postmenopause patients(71.8%) than that in premenopause ones(89.5%).(3)TRPV6-positive expression was higher in the cases of cervical involvement(89.3%) than the cases of non-involvement(67.7%).Conclusion:The expression of TRPV6 in endometrial cancer is lower than that in the normal endometrium,and TRPV6-positive tissues are tend to have more cervical involvement.It suggests that calcium channel protein TRPV6 has relationship with endometrial cancer.更多还原
