TGF-β1及CTGF在PCOS大鼠的表达及罗格列酮的干预作用

【摘要】 目的:探讨转化生长因子-β1(TGF-β1)及结缔组织生长因子(CTGF)在P-COS大鼠卵巢中的表达及罗格列酮的干预作用。方法:随机将60只雌性SD大鼠分为对照组、模型组和治疗组。模型组和治疗组大鼠在22日龄时用皮下注射脱氢表雄酮(DHEA)法建立PCOS动物模型,治疗组从注射DHEA时起予以罗格列酮3mg/(kg.d)灌胃。第20天处死大鼠,HE染色光镜下观察大鼠卵巢的病理结构;透射电镜观察细胞超微结构;微粒子酶免分析法测定血清激素水平的变化;用免疫组织化学法测定大鼠卵巢组织TGF-β1、CTGF蛋白表达及定位,并用病理图像分析仪分析图像。结果:大鼠血清激素水平,卵巢的病理结构变化以及细胞超微结构验证模型建立成功。模型组卵巢组织TGF-β1、CTGF蛋白表达显著高于对照组(P<0.05,P<0.001),罗格列酮治疗后,卵巢中CTGF、TGF-β1表达水平均较模型组显著降低(P<0.05,P<0.001)。模型组血清TGF-β1、CTGF表达也显著高于对照组(P<0.05,P<0.001),罗格列酮治疗后,血清中二者水平均较模型组显著降低(P<0.05,P<0.001)。结论:TGF-β1及CTGF协同参与了PCOS大鼠卵巢纤维化的发生、发展过程。罗格列酮可延缓卵巢纤维化的发生发展,其与降低PCOS大鼠的CTGF、TGF-β1的表达有关。

【Abstract】 Objective:To explore the expressions of transforming growth factor-β1(TGF-β1) and connective tissue growth factor(CTGF) in ovarian tissue of polycystic ovary syndrome(PCOS) rat and the intervention effect of rosiglitazone(RGZ)on them.Methods:Sixty female SD rats were randomly divided into 3 groups:Control group,model group and treatment group.Model group and treatment group were established PCOS model by subcutaneous injection Dehydroepiandrosterone(DHEA) from 22 days of age,meanwhile treatment groups received RGZ 3 mg·kg-1·d-1 by intragastric administration.They were killed after 20 days.Serum hormone values of the rats were tested by microparticle enzyme immunoassay,the pathohistology of rats’ ovarian structure was observed by hematoxylin-eosin(H&E) staining,The ovarian ultrastructure was investigated by transmission electron microscope.and the expressions of TGF-β1 and CTGF were detected and evaluated by immunohistochemistry and image analysis system.Results:The results of serum level,and the pathological-histology,the ultrastructure of ovary showed that the model was established successfully.Compared with control groups,the expression of TGF-β1 and CTGF protein in ovaries increased markedly in model groups(P<0.05,P<0.001).After treatment with RGZ,the expressions of TGF-β1 and CTGF protein in rats ovaries decreased significantly when compared with model groups(P<0.05,P<0.001).Also the expression of TGF-β1 and CTGF in rats serum increased markedly in model groups(P<0.05,P<0.001),and after treatment with RGZ,the expressions decreased significantly in treatment groups(P<0.05,P<0.001).Conclusions:TGF-β1 and CTGF may play important roles in the development of ovary fibrosis in PCOS.RGZ can postpone the development of ovary fibrosis through decreasing the levels of TGF-β1 and CTGF.
