(R)-5-扁桃酰基-3-对甲基哌嗪苯甲酰基-4,6-二氢吡咯[3,4-c]吡唑的合成工艺改进
Improved synthesis of (R)-N-(5-(2-methoxy-2-phenylacetyl)-1,4,5,6-tetrahydropyrrolo[3,4-c] pyrazol-3-yl)-4-(4-methylpiperazin-1-yl)benzamide
【摘要】 以甘氨酸为起始原料,经Micheal反应、酯化、氨基保护、环合、得3-氨基-4,6-二氢吡咯并[3,4,c]吡唑-5(1H)-叔丁氧羰基,再经酰化、脱boc保护,生成的化合物与扁桃酸酰化、脱保护得(R)-N-5-(2-甲氧基-2-苯乙酰)-3-([4-(4-甲基哌嗪)苯甲酰基]氨基)-4,6-二氢吡咯[3,4-c]吡唑,总收率为24.5%。
【Abstract】 (R)-N-(5-(2-methoxy-2-phenylacetyl)-1,4,5,6-tetrahydropyrrolo[3,4-c]pyrazol-3-yl)-4-(4-methylpiperazin-1-yl)benzamide was synthesized from glycine via Micheal reaction,esterification,protection of amino,cyclization to give tert-Butyl-3-Amino-4,6-dihydro-1H-pyrrolo[3,4-c]pyrazole-5(1H)-carboxylate,which followed by acylation,deprotection of boc,acylation with mandelic acid and deprotection with an overall yield of about 24.5%.
