氟化钠对体外培养大鼠心肌细胞培养液中NO浓度影响

【摘要】 目的探讨氟化物对体外培养心肌细胞的毒性作用机制。方法建立体外培养心肌细胞模型,应用生物化学方法研究不同剂量氟化钠处理后,心肌细胞搏动与停搏情况及培养液中NO浓度的变化。结果心肌细胞的停搏率与氟离子(F-)剂量间存在明显的正相关关系(r=0.9688),F-的半数停搏浓度为8.05mg/l。D组心肌细胞染毒前、后培养液中NO浓度改变有统计学意义(P<0.05)。结论高剂量的氟化钠可能诱导心肌细胞合成大量的NO,其毒作用表现为心肌细胞的收缩力减弱,直至停搏。

【Abstract】 Objective To investigate the mechanism of toxic effects on myocardial cells cultured in vitro with sodium fluoride(NaF). Methods The model of myocardial cells cultured in vitro and the methods of biochemistry technology were applied to observe the content of nitric oxide (NO)during heart beating as well as beat-arrested myocardial cells exposed to NaF. Results After treated with NaF for 24h,the myocardial cells in each group had the beat-arrested cells. With the increasing of the NaF dosage,the ratio of beat-arrested cells increased,and showed a positive correlation (r=0.9688). The concentration of NO in group D increased after treated with NaF for 24h (P<0.05 ). Conclusion High dosage of NaF may induce the release of large quantity of NO by myocardial cells leading to the decrease of contractile force of myocardial cells and stopping beating.
