α-干扰素对人肺腺癌A549细胞生物学行为的影响

【摘要】 目的:研究IFN-α对人肺腺癌A549细胞株克隆形成、迁移和侵袭能力以及细胞凋亡的影响。方法:用不同浓度(100、500、1 000、2 000 U.ml-1)IFN-α处理人肺腺癌A549细胞(实验组),以未经药物处理细胞作对照(对照组),平板克隆形成实验观察两组细胞克隆形成能力的差异,Transwell小室实验观察两组细胞迁移和侵袭能力的差异,流式细胞仪观察两组细胞凋亡水平的差异。结果:IFN-α能够明显降低人肺腺癌A549细胞克隆形成、迁移和侵袭能力,诱导细胞的凋亡,而且在一定范围内与药物浓度成正相关(P<0.05)。结论:IFN-α在非小细胞肺癌临床治疗中可能有潜在价值。

【Abstract】 Objective:To study the influence of interferon-α(IFN-α) on the colony formation,migration,invasion and apoptosis of human lung adenocarcinoma A549 cell line.Methods:Human lung adenocarcinoma A549 cells were treated with a concentration of 100,500,1 000,2 000 U·ml-1IFN-α,respectively.The ability of cell clone formation was detected by the plate colony forming assay.The cell migration and invasion ability were observed in Transwell chamber.The apoptosis was analyzed by flow cytometry.Results:The ability of cell colony formation,migration and invasion were significantly reduced.IFN-α coulde induce cell apoptosis in a concentration-dependent manner(P<0.05).Conclusions:IFN-α may have potential value in non-small cell lung cancer treatment.
