星点设计-效应面法优选白藜芦醇固体脂质纳米粒处方

【摘要】 目的:采用星点设计-效应面法优选白藜芦醇固体脂质纳米粒的处方。方法:采用星点设计-效应面优化法筛选处方,以白藜芦醇包封率及载药量作为评价指标,考察脂质用量、表面活性剂用量、药物量对评价的影响。采用热高压均质法制备白藜芦醇固体脂质纳米粒。结果:热高压均质工艺能有效制备固体脂质纳米粒,优选的最佳处方为单硬脂酸甘油酯用量为4.5%、泊洛沙姆用量为6%、白藜芦醇投药量为0.45%,OD值与理论值偏差-4.12%。结论:星点设计-效应面法能有效优选RES-SLN处方。

【Abstract】 Objective:To optimize the formula of resveratrol-loaded solid lipid nanoparticles(RES-SLN).Methods:RES-SLN was prepareded by hot-high pressure homogenization technique.Central composite design/response surface methodology was applied to screen RES-SLN prescription.Independent variables were lipid content,Poloxamer 188(F68)content and RES content.Results:The optimum prescription was the follow:Glycerol Monostearate(GMS)4.5%,F68 6%,RES 0.45%.The particle size of RES-SLN was about 100nm.The bias was-4.12%.Conclusion:Cental composite design-response surface metogology is successflully used to optimize the formulation of RES-SLN.
