高压培养促进人血管内皮细胞和人单核细胞的粘附

【摘要】 目的探讨体外高压培养对人血管内皮细胞粘附功能的影响,为高血压和血管动脉粥样硬化的发病关系提供实验依据。方法体外培养血管内皮细胞,分别以不同的压力(0、120、150、180、210 mmHg)培养细胞,细胞计数法测定内皮细胞与单核细胞的粘附率。结果高压培养明显增加内皮细胞和单核细胞的粘附率,180 mmHg高压培养细胞较大气压力(0 mmHg)培养24 h粘附率增加约3.6倍;但MG132预处理抑制了180 mmHg高压培养诱导的内皮细胞与单核细胞的粘附率。结论高压培养促进血管内皮细胞的粘附功能。

【Abstract】 Objective Hypertension plays an important role in the development of Atherosclerosis.The object of this study is to observe the effect of static pressure on the human vascular endothelial cell adhere. Methods Cell counting under microscope detected the adhesion rate between endothelial cells Human-12 and THP-1 cells after treated with different static pressure 0,120,150,180,210 mmHg. Results Static pressure up-regulated the adhesion rates between HUVEC-12 and THP-1 cells.It showed about 3.6-fold increase after treatment with high pressure 180 mmHg for 24 hrs.However,pre-treatment with MG132 decreased the induction of high pressure on adhesion rate between HUVEC-12 and THP-1 cells. Conclusion Static high pressure culture may induce the vascular endothelial cells adhesion function probably through the nuclear factor-κB pathway.
