胃肠道腺癌组织中HIF-1α、COX-2及Bax蛋白的表达及意义

【摘要】 目的探讨缺氧诱导因子1(HIF-1α)、环氧化酶-2(COX-2)及Bax在胃肠道癌组织中的表达及其临床意义。方法以经病理证实的胃肠道腺癌组织37例为胃肠道腺癌组,以其胃肠道腺癌淋巴结转移组织37例作为淋巴结转移癌组,正常胃肠道黏膜组织37例作为正常胃肠道黏膜组。采用链霉素-生物素(S-P)免疫组化技术,分别测定3组癌组织中HIF-1α、COX-2及Bax的表达。采用Wilcoxon秩和检验进行组间比较。结果胃肠道腺癌、胃肠道腺癌淋巴结转移组织中HIF-1α、COX-2及Bax蛋白阳性表达率均明显高于正常胃肠道黏膜组织(P<0.01);胃肠道腺癌组织淋巴结转移的癌组织及胃肠道腺癌组织中HIF-1α、COX-2及Bax蛋白阳性表达率间差异无统计学意义(P>0.05)。结论HIF-1α、COX-2及Bax蛋白在胃肠道腺癌及其淋巴结转移癌的组织中表达水平高于正常胃肠道黏膜,其表达水平与肿瘤的发生、发展、浸润、转移相关,检测上述标志物有一定的临床及病理意义。

【Abstract】 Objective To investigate the expression of hypoxia-inducible factor-1α,cyclooxygenase-2 and Bax in gastrointestinal adenocarcioma and its clinical significance. Methods The 37 cases of gastrointestinal adenocarcioma identified by pathology were used as gastrointestinal adenocarcioma group,37 cases gastrointestinal adenocarcioma with lympho node metastasis were used as lymph node metastasis group and 37 cases of normal gastrointestinal mucous membrane were used as control group. Immunohistochemistry SP method was used to examine the expression of HIF-1α,COX-2 and Bax in gastrointestinal adenocarcioma,normal gastrointestinal mucous membrane and metastasis lymph node of gastrointestinal adenocarcioma. The comparison among groups was performed by Wilcoxon sum of ranks. Results The positive expression rates of HIF-1α,COX-2 and Bax both in gastrointestinal adenocarcioma group and lymph node metastasis group were significantly higher than those in control group( P < 0. 01). There was no significant difference in the positive expression rate of HIF-1α,COX-2 and Bax between gastrointestinal adenocarcioma group and lymph node metastasis group( P > 0. 05). Conclusion The expression of HIF-1α,COX-2 and Bax protein in gastrointestinal adenocarcioma and metastasis lymph node tissue is higher than that in normal gastrointestinal mucous membrane. They may play an important role in the pathogenesis,progression,invasion and metastases of the gastrointestinal adenocarcioma. Detemination of HIF-1α,COX-2 and Bax is helpful for the diagnosis and therapy of gastrointestinal adenocarcioma.
