海洛因急性中毒大鼠血脑屏障的改变

【摘要】 目的:探讨海洛因急性中毒血脑屏障通透性的变化。方法:雌性SD大鼠20只,随机分为生理盐水对照组和海洛因模型组。模型组采用快速递增法大鼠皮下注射海洛因,人为建立海洛因急性中毒动物模型。对照组用同样方式注射不含海洛因的生理盐水。模型组注射纳洛酮催瘾,动物出现戒断症状,应用Maldonado评分标准判断成瘾强度。2组大鼠麻醉后经右股静脉给予示踪剂伊文思蓝(Evans Blue,EB),用荧光显微镜观察血脑屏障的完整性。结果:海洛因急性中毒模型组大鼠荧光显微镜下额叶、顶叶、海马、间脑、小脑、脑干各部位红色荧光光斑明显增多,每10个高倍镜视野EB光斑数与对照组比较,差异有统计学意义(P<0.01)。结论:大鼠海洛因急性中毒后血脑屏障通透性增大,破坏了脑组织内环境的稳定,阻碍了脑组织与外周组织的物质及信息传递,导致脑细胞水肿、变性、坏死。

【Abstract】 Objective:To discuss the blood-brain barrier permeability changes in heroin acute poisoning rats.Methods:Twenty healthy adult female SD rats were randomly divided into saline digital control group and model group of heroin.Applying the method of quickly increasing,we inject heroin on subcutaneous to artificially establish animal model of heroin acute poisoning.The control group was established by the same way by injecting saline.Injecting naloxone to remind addiction and judge addiction strength after withdrawal symptoms occurrence by Maldonado score.Two groups of rats were anesthetized by the right femoral vein for tracer Evans blue to observe the integrity of blood-brain barrier with fluorescence microscope.Results:Model of acute poisoning of heroin in rat brain frontal lobe,parietal lobe,hippocampus,diencephalon,cerebellum,brainstem different parts of the red fluorescent spots increased significantly and high magnification view of 10 the number of EB spot differences between the control group(P<0.01).Conclusion:Heroin acute poisoning increases the permeability of blood-brain barrier,damages brain tissue homeostasis,hinders the brain and peripheral tissue of the material and information transfer,leads to brain cell swelling,degeneration,necrosis.
