【摘要】 目的:研究"左归丸"不同配伍对骨髓源成体干细胞(adult stem cells,ASC)定向分化的调控作用。方法:以免疫组化法鉴定"左归丸"撤药各方诱导ASC定向分化的干/祖细胞为指标,观察左归丸的配伍规律。结果:从方中撤出牛膝后Ⅱ型胶原表达明显降低;撤出山药碱性磷酸酶活性降低。撤出枸杞、山茱萸神经元烯醇化酶(NSE)表达降低,但撤出熟地后NSE表达明显升高;撤出枸杞、熟地后Ⅷ因子表达降低,但撤出山茱萸后Ⅷ因子表达明显升高。结论:牛膝可诱导ASC向软骨细胞方向分化;山药可诱导ASC向成骨细胞方向分化。枸杞、山茱萸可诱导ASC向神经元方向分化,但方中熟地可能具有抑制ASC向神经元方向分化的作用;枸杞、熟地可诱导ASC向内皮细胞方向分化,但山茱萸可能具有抑制ASC向内皮细胞方向分化的作用。结果提示,左归丸配伍规律在诱导干细胞分化与抑制分化方面起着重要的调控作用。更多还原

【Abstract】 Objective:To investigate the controlled effect of Zuo-Gui Pill compatibility on directional differentiation of the adult stem cells(ASC).Methods:immunohistochemical method was used to identify the withdrawal herb which induct ASC’s directional differentiation’s stem cell in the Zuo-Gui Pill,then,to observe the compatibility rule of Zuo-Gui Pill.Result:the expression of theⅡcollagen was reduced obviously after stopping using Niu Xi,the activity of alkaline phosphatase was reduced when Shan Yao is stopped.The expression of neural enolase was reduced when Gou Qi and Shan Zhu Yu were withdrawal,however,it was increased when Shu Di was withdrawal.And the expression of VIII factor was lower after the Gou Qi and Shu Di were stopped.However,at the same time,it was increased obviously when Shan Zhu Yu is stopped.Conclusion:Niu Xi can induce ASC differentiate to chondrocyte,Shan Yao can induce it to osteoblast,Gou Qi and Shan Zhu Yu induce it to neuron,but Shu Di perhaps inhibit ASC’s differentiate to neuron.Gou Qi and Shu Di induce ASC to endothelial cell when Shan Zhu Yu inhibits it to endothelial cell.As a result,it is so important of the Zuo-Gui Pill compatibility on controlling differentiation of the bone marrow-derived adult stem cells.更多还原
