湖南省40株狂犬病毒株N基因的序列分析
Analysis on N gene sequence of 40 rabies virus strains in Hunan province
【摘要】 目的研究湖南省40株狂犬病毒株N基因序列特征及其遗传进化关系。方法采用RT-PCR法从家犬脑组织标本检测狂犬病毒RNA,并对扩增产物N基因片断进行测序。采用DNAStar软件包中MegAlign将所得测序结果与湖南省往年20株毒株N基因进行核苷酸、氨基酸序列同源性比对分析,并以Neighbor-Joining法构建系统发生树。结果40株毒株均为基因1型,N基因核苷酸序列同源性在88.5%~100%(中位数97.9%)间,编码的氨基酸序列同源性为98.3%~100%(中位值99.6%)。系统发生分为3个组群,具有一定的地域性。结论湖南省40株狂犬病毒株间N基因变异较小,稳定性较高。
【Abstract】 To analyze the characteristics of the N-gene sequence in 40 strains of rabies virus isolated in Hunan province and their phylegenetic correlation among these strains,the viral RNA of rabies virus from brain tissues of domestic dogs was assayed by RT-PCR and the N-gene fragment the amplified product was sequenced later on.MegAlign bio-software attached to DNA Star package was further employed for sequence alignment of both nucleotides and conducted amino acids between the results of the investigated rabies virus strains and those of 20 strains previously isolated in Hunan province.The Neighbor-Joining method was subsequently conducted to build up the phylogenetic tree.It was found that all these 40 rabies viruses belonged to genetype 1.and they enjoyed nucleotide identitues of 88.5%-100%(medium at 97.9%),and those of the conducted amino acid identities were 98.3%-100%(medium at 99.6%).The analysis of phylogenetic tree was demonstrated to be genetically divided into three groups,indicating that the virus presented a unique characterization in geographic distribution.From these observations,it is evident that the nucleotide and amino acid sequence of N genes appeared to be stable and little variance was found among the 40 strains of rabies virus isolated in Hunan province.
