棉籽粕提取分离蛋白工艺的优化研究
Optimizing Extraction of Cottonseed Isolated Protein
【摘要】 以棉籽粕为原料,对几种不同的棉籽蛋白提取以及脱毒方法进行了比较,并利用正交旋转旋转组合设计优化棉籽蛋白的提取工艺。结果表明,热碱法既能保证较高的蛋白得率和含量,还有很好的脱酚效果;通过四元二次通用旋转组合设计试验,得到最佳工艺条件为pH 10.71,温度60.2℃,时间1.07 h,液固比16.7∶1,其蛋白得率≥60%,蛋白质含量≥90%。
【Abstract】 Several different methods for extraction and degossypol of cottonseed protein were compared with cottonseed as raw material.The extraction conditions were also optimized using orthogonal rotary unitized design.Results show that the hot aqueous alkali method can ensure products with high protein yield and protein content,and with good degossypol effect.The obtained optimal conditions are pH 10.71,temperature 60.2 ℃,time 1.07 h and the ratio of liquid to solid 16.7∶1.The protein yield and product protein content are over 60% and 90%,respectively.
