嗅成鞘细胞提取液对阿尔茨海默病脑移植的影响

【摘要】 目的探讨嗅成鞘细胞提取液对阿尔茨海默病(AD)脑移植的影响。方法SD大鼠双侧海马注射Aβ1-40,建立AD大鼠模型。实验动物分为四组:正常对照组、空白对照组、人工脑脊液移植组、嗅成鞘细胞OECs提取液移植组,每组10只。两移植组分别加用人工脑脊液、OECs提取液培养胆碱能神经元,并移植到AD模型大鼠。运用行为学测试、NADPH-d组化、刚果红组织学染色结合积分吸光度测定等技术,观察、比较各组AD模型大鼠移植2个月后学习记忆能力、一氧化氮合酶(NOS)阳性神经元数以及老年斑的变化。结果①行为学测试:人工脑脊液移植组、OECs提取液移植组大鼠达到学会标准所需训练次数(TN)、错误反应次数(EN)、潜伏期(D)、全天总反应时间(TRT)少且短于空白对照组(P<0.05);OECs提取液移植组更为明显,与人工脑脊液移植组之间有显著差异(P<0.05)。②NADPH-d组化反应:人工脑脊液移植组、OECs提取液移植组NOS阳性神经元数较空白对照组明显增多(P<0.05),且前两组之间有显著性差异。③刚果红染色:SP积分吸光度测定,三组间均有显著性差异(P<0.05),OECs提取液移植组SP-IA值最小。结论OECs提取液能明显提高AD脑移植的效果。

【Abstract】 Objective To explore the effect of distilled fluid of olfactory ensheathing cells(DFOECs)transplantation on Alzheimer’s disease(AD).Methods The AD model rats were made by injecting Aβ1~40 into double hippocampus.The rats were divided into normal control,blank control,artificial cerebrospinal fluid(ACSF)transplanted and DFOECs transplanted groups randomly,and there were eight rats each group.Cholinergic neuron was cultured with ACSF and DFOECs respectively in two transplanted groups.Two months later,the ability of learning and memory,nitric oxide synthase(NOS)positive neurons and senile plaques were observed by Y-labyrinth test,NADPH-d histochemical and Congo red histological staining.Results ①Y-labyrinth test:training number(TN),error number(EN),delitescence(D),total reaction time(TRT)of ACSF and DFOECs transplanted groups obviously decreased compared to those of blank control group(P<0.05),especially DFOECs group(P<0.05).② NADPH-d histochemical staining:the total number of NOS positive neurons of ACSF and DFOECs transplanted groups were higher than those of blank control group(P<0.05),there was significant difference between the two transplanted groups(P<0.05).③ Congo red histochemical staining:compared to normal control group,the other three groups had significant change in SP-IA(P<0.05),DFOECs of transplanted group was the smallest.Conclusions DFOECs can greatly improve the effect of cholinergic neuron transplanted for treatment of AD rat.
