实验性脑积水幼鼠脑脊液中一氧化氮及谷氨酸浓度的变化

【摘要】 目的探讨实验性脑积水幼鼠脑脊液中一氧化氮(NO)和谷氨酸(Glu)浓度的动态变化。方法SD幼鼠40只,随机平分为A、B、C实验组和对照组共4组,用显微外科技术向枕大池内注入25%白陶土混悬液,分别在注射后第2(A组)、3(B组)、4周(C组)后行MRI检测,鉴定脑积水形成情况;之后24h内取脑脊液,用硝酸还原酶法测NO浓度、高效液相色谱仪检测Glu浓度。对照组幼鼠于枕大池内注入生理盐水,用同样方法检测。结果实验组21只幼鼠成功诱发脑积水。与对照组相比,实验组脑脊液中NO和Glu浓度显著增加(P<0.01)。结论脑脊液中高水平的NO和Glu可能参与了实验性脑积水幼鼠的病理损伤过程。

【Abstract】 Objective To explore the Changes in nitric oxide (NO) and glutamic acid(Glu) levels in the cerebrospinal fluid (CSF) of young rats with experimental hydrocephalus and their meaning. Methods Forty young SD rats were randomly divided into four groups of 10 rats each, i.e. experiment groups A, B and C, and control group. The hydrocephalus was induced by injecting 25% kaolin solution into the cisterna magna in the rats of all the experimental groups. The injection of the same volume of 0.9% sterile saline as the kaolin solution injected in the experiment groups into the cisterna magna was performed. MRI was performed 2 (group A), 3 (group B) and 4 (group C) weeks respectively after the injection of Kaolin solution in all the experimental groups to observe the hydrocephalus. The CSF were removed out within 24 hours after the MRI examination. NO and Glu levels in the CSF were detected respectively by the nitrite/ nitrate assay and high performance liquid chromatography in all the groups. Results The hydrocephalus was successfully induced in 21 mice, of whom, 8 were in the experimental group A, 8 in the group B and 5 in the group C. The NO and Glu levels of the CSF in rats with hydrocephalus were significantly higher than that in the rats of the control group (P<0.01). Conclusion It is suggested that the upregulation of the levels of NO and Glu in the CSF may take part in the pathological injury induced by the hydrocephalus.
