黄芪、莪术配伍方对胃癌cox-2、pparγ及转移相关因子表达的影响
The influence of compound of Radix Astragali and Rhizoma curcumae on the expression of cox-2,pparγand other metabasis-correlating factors in gastric cancer
【摘要】 目的:观察黄芪、莪术配伍方对胃癌细胞MKN45中COX-2、PPARγ、MMP-2、VEGF表达的影响。方法:以黄芪、莪术配伍作用于MKN-45细胞,并设塞莱昔布组(cox-2抑制剂)、罗格列酮组(pparγ激动剂)和空白组相对照。通过ELISA法检测VEGF水平的变化,用RT-PCR以及Western blot方法检测加药后胃癌细胞cox-2、pparγ、mmp-2基因及COX-2蛋白表达的变化。结果:中西药组对VEGF表达的抑制效应起始于48h,以塞菜昔布组和中药组作用显著。各药物对COX-2蛋白、cox-2mRNA和mmp-2mRNA均有抑制作用,对pparγmRNA有促表达作用。其中对COX-2的抑制以塞莱昔布组和中药组最明显,且呈时间依赖性。对MMP-2的抑制作用起始于48h,以塞莱昔布组和中药组较明显。罗格列酮组和中药组对pparγ的促表达作用最明显。结论:说明益气活血疗法对cox-2及其上下游基因有明显调控的作用,中药抗肿瘤作用可能是对肿瘤基因多靶点作用的结果。
【Abstract】 Objective:To survey the effect of Chinese herb medicines compound of Radix Astragali and Rhizoma curcumae on the expression of COX-2,PPARγ,MMP-2,VEGF in gastric cancer cell MKN-45.Methods:The decoction of Radia Astragali and Rhizoma curcumae was treated on gastric cancer cell MKN-45,contrasting with two positive drugs celecoxib(COX-2 inhibitor) and rosiglitazone (pparγexcitant).VEGF was detecded by ELISA.Cox-2 mRNA,pparγmRNA,mmp-2 mRNA were examined through reverse transcription-polymerase chain reaction and Western blot method.Results: All the examined groups can depress VEGF after 48h,the celecoxib group and herb medicine group possess more strong activity.All the groups can inhibit cox-2,cox-2 m RNA,mmp-2 mRNA and promote the expression of pparγmRNA.Among which,the celecoxib and Chinese herbs had best sup- pressant and influence on cox-2.Conclusion:The Chinese herb medicines,Radix Astragali and Rhizoma curcumae had apparent regulatory-effect on cox-2 and its up-down stream gene.Anti-tumor action of Chinese herb medicines may be through effecting multi-target of tumor gene.
