吗啡依赖大鼠海马CA1区神经元超微结构的时序性变化
Sequential changes of neuronic ultrastructures in hippocampus CA1 of morphine dependent rats
【摘要】 目的研究高、低吗啡条件性位置偏爱(CPP)易感性大鼠海马CA1区神经元超微结构的时序性变化,为阿片依赖提供形态学依据。方法160只雄性SD大鼠随机分为实验组(n=130)和对照组(n=30)。实验组按剂量递增法腹腔注射吗啡,每日2次,起始剂量为每次5 mg.kg-1,逐日递增每次5 mg.kg-1,共10 d。对照组同期注射相同体积的生理盐水。2组动物均进行CPP测评。实验组大鼠经过CPP测评后再次分为高、中、低偏爱组,其中中偏爱组淘汰,高、低偏爱组大鼠分别进入以下研究:分别在末次吗啡注射后3 h、3 d、14 d处死大鼠,制作电镜样本。应用透射电镜对2组大鼠海马CA1区进行观察,并与对照组进行比较。结果各个时点高、低偏爱组大鼠与对照组相比,神经元核膜节段性不清、断裂。核内染色质聚集成团、边集;线粒体肿胀变圆、内有空泡形成、嵴排列紊乱、嵴断裂、畸形线粒体出现;粗面内质网扩张、脱颗粒;高尔基体肿胀;上述病理改变不随时间明显加重,且高偏爱组的病理改变较低偏爱组严重。结论吗啡处理可导致海马CA1区神经元超微结构发生病理改变,这种病理改变不随时间明显加重,且具有个体易感性。
【Abstract】 Objective To investigate sequential changes of neuronic ultrastructure in hippocampus CA1 in high and low conditioned place preference(CPP) rats at different time points after chronic morphine treatment.Methods One hundred and sixty male SD rats were randomly divideded to experiment group(130 cases) and control group(30 cases).The rats in experiment group were intraperitoneally injected with morphine,twice a day for ten days,in an ascending dosage schedule,the initial dose was 5 mg·kg-1,the progressive dose was 5 mg·kg-1.The rats in control group were intraperitoneally injected with the same voluminal normal saline at same time.The rats in experiment group were divided into high preference group,middle group and low preference group according to the numerical value of the CPP.The middle group was rejected,the rate in high preference group and low preference group were sacrificed at the time of 3 hours,3 days and 14 days after the last injection.The hippocampus CA1 were removed and prepared for electron microscope specimen.The ultrastructural pathologic change of neurons was examined by transmission electron microscope.Results Compared with control group,the changes of neuronic ultrastructure in hippocampus CA1 in high preference group and low preference group were more severes.The nuclear membranes were blurry.The nuclear chromatin was aggregate.The mitochondria was oncootic and vacuolate,mitochondria strutting was disheveled.The rough endoplasmic reticulum and dictyosome were dilative.The changes didn′t become more severe along the time.Conclusion Morphine can induce severe ultrastructural pathologic changes in hippocampal CA1 area,and the changes don′t become more severe along the time,and these changes have individual susceptibility.
