【摘要】 目的比较食管鳞癌和宫颈鳞癌组织中9号染色体相同区域相同位点等位基因杂合性缺失(LOH)率,探索与二者共同相关的抑癌基因。方法采用显微切割、PCR、变性聚丙烯酰胺凝胶电泳、AgNO3染色等技术,检测45例食管鳞癌和46例宫颈鳞癌组织中的9号染色体上7个微卫星位点的LOH。结果食管鳞癌总LOH率为25.8%(55/213),7个微卫星位点的LOH率从大到小依次为D9S753(42.9%)、D9S303(38.5%)、D9S171(35.7%)、D9S162(36.4%)、D9S242(16.7%)、D9S1748(16.7%)、D9S43(0)。宫颈鳞癌中总LOH率为25.2%(54/214),7个微卫星位点的LOH率从大到小依次为D9S171(40.7%)、D9S43(33.3%)、D9S162(31.4%)、D9S242(23.7%)、D9S303(17.2%)、D9S753(16.7%)、D9S1748(12.9%)。结论食管鳞癌和宫颈鳞癌中存在相同的高频率的LOH位点,9号染色体上可能存在与二者发生发展共同相关的抑癌基因。更多还原

【Abstract】 Objective To compare the rates of loss of heterozygous(LOH)of allelomorphic gene on chromosome 9 at the same regions and sites between esophageal squamous cell carcinoma and squamous carcinoma of the cervix,and to explore their related tumor suppressor genes in common. Methods LOH was detected in 45 cases of esophageal squamous cell carcinoma and 46 cases of squamous carcinoma of the cervix using microdissection,polymerase chain reaction,denaturing polyacrylamide gel electrophoresis and silver nitrate staining. Results The total frequency of LOH in esophageal squamous cell carcinoma was 25.8%,and rates of LOH were 42.9% at D9S753,38.5% at D9S303,35.7% at D9S171,36.4% at D9S162,16.7% at D9S242,16.7% at D9S1748,0 at D9S43.The total frequency of LOH in squamous carcinoma of the cervix was 25.2%,and rates of LOH were 40.7% at D9S171,33.3% at D9S43,31.4% at D9S162,23.7% at D9S242,17.2% at D9S303,16.7% at D9S753,12.9% at D9S1748. Conclusion The identical high frequency LOH sites exist in esophageal squamous cell carcinoma and squamous carcinoma of the cervix.The tumor suppressor genes related to the development of the two squamous carcinomas in common maybe exist on chromosome 9.更多还原
