产漆酶木腐真菌的筛选、鉴定及部分酶学性质

【摘要】 综合菌体培养方式和培养基组成对菌株产漆酶性能的影响,对68株木腐菌进行了筛选,获得了1株适合液态发酵产漆酶的漏斗多孔菌A08,酶学性质研究表明,漆酶反应最适温度为60-65℃,最适pH为5.5,60℃时酶活半衰期大于45min,20℃酶活性在pH3.0-9.0稳定,漆酶活性受Cl-、Ca2+、Fe2+抑制,Mn2+激活,K+、Mg2+、Zn2+、Cu2+等对酶活性没有显著影响。

【Abstract】 Based on the effects of culture method and medium composition upon productivity of laccase from Wood-rot Fungi,a strain with relative higher laccase-producing ability in liquid culture was screened from 68 strains of wood-rot fungi,and identified as Polyporus arcularius.Studying on enzymatic properties revealed that the optimum temperature and pH of the laccase reacted with guaiacol were 60-65℃ and pH 5.5,respectively. The half-life of the laccase was more than 45 min at 60℃. The enzyme activity was stable within pH range of 3.0-9.0 under 20℃,was strongly inhibited by Fe2+,and moderately by Cl-,Ca2+,while enhanced by Mn2+. Wheres other ions like K+,Mg2+,Zn2+,Cu2+ had no evident effects on it within the range of experimental concentration.
