产D-塔格糖菌株的筛选与鉴定

【摘要】 从土壤、泡菜汁、牛奶等样品中筛选获得2株高产D-塔格糖的菌株,根据16S rDNA序列比对并结合菌株的形态特征、生理生化特征分析,鉴定其分别为发酵乳杆菌和植物乳杆菌,且命名为Lactobacillus fermentumNXTag-1和Lactobacillus plantarum NXTag-2。菌株NXTag-1和NXTag-2表现出不同的产L-阿拉伯糖异构酶的能力,其最高酶活分别为5.93U/mL和5.05U/mL,以80g/LD-半乳糖为底物,分别产30g/L和26g/L的D-塔格糖。

【Abstract】 Two D-tagatose-producing strains,NXTag-1 and NXTag-2,were isolated from the samples of soil,pickle and milk.Based on the sequence analysis of 16S rDNA and a series of morphological and biochemical characteristics,NXTag-1 was identified as Lactobacillus fermentum,NXTag-2 was identified as Lactobacillus plantarum.The maximum of L-arabinose isomerase activity of NXTag-1 and NXTag-2 reached 5.93 U/mL and 5.05 U/mL,the yield of D-tagatose were 30 g/L and 26 g/L(80 g/L D-galactose),respectively.
