几种因子对鞍带石斑鱼精子活力的影响
Effect of Several Factors on Motility of Sperm of Epinephelus lanceolatus
【摘要】 研究不同梯度的盐度、pH、温度及低温保存时间对鞍带石斑鱼精子活力的影响。结果表明:鞍带石斑鱼精子活力的适宜盐度范围为27.5~35.0,最适盐度为32.5,此时精子的快速运动时间和寿命分别为13.2min和14.8min;最适pH为8.5,精子的快速运动时间及寿命最长,分别达12.7min和17.3min;温度范围为27~31℃,以29℃时快速运动时间最长,达16.6min;在0~4℃冰箱保存6h后鞍带石斑鱼精子活力未显著降低,其中保存前精子快速运动时间为18.4min,保存6h后为18.2min,保存120h后仍有活力,精子快速运动时间为2.3min,精子激活率为3.3%。以x为保存时间,快速运动时间y和寿命y′符合线性方程y=-0.8334x+19.164(R2=0.9856),y′=-1.1358x+27.8(R2=0.9573)。
【Abstract】 Salinity,pH,temperature and preservation time were used to assess the sperm viability of Epinephelus lanceolatus by means of different scales of those factors,and as the experiments on process,the optimal value of the front factors set as the following experiment conditions. The optimal motility was obtained at salinity between 27.5-35.0,with the longest viability at 32.5 recording the motility time and duration of 13.2 and 14.8 min,respectively. With respect to pH,the longest time of fast motility and duration was acquired,when the value was 8.5,with the motility time and duration of 12.7 and 17.3 min,respectively. A maximum motility time of 16.6min was recorded in 29 ℃,with the best suitable temperature ranging 27 -31℃. Of all the three factors,the sperm optimal viability requirement was the same as that of the breeding conditions. After preserved at temperature 0 -4 ℃ for 6 hours the motility time of sperm was 18.2 min,which was not significantly reduced compared with the initial time 18.4min. After 120 hours stored at the same temperature,the motility time of sperm was 2.3 min and the activation rate was 3.3%. The viability debased following the preservation time calculated,suggesting the sperm should be used as soon as possible after preserved. The motility time ( y ) and duration ( y’ ) fitted the equations:y = -0.8334 x + 19.164,R2 = 0.9856; y’ = -1.1358 x + 27.8,R2 = 0.9573.
