逍遥散对慢性应激大鼠HPA轴负反馈调节功能的影响
Effect of Xiaoyaosan on HPA Axis Negative Feedback Regulation Function of Rats With Chronic Stress
【摘要】 目的:研究中药复方逍遥散及糖皮质激素受体(GR)阻滞剂米非司酮(RU-38486)对慢性应激大鼠下丘脑-垂体-肾上腺(HPA)轴的负反馈调节功能。方法:健康雄性Wistar大鼠随机分为5组,模型组、地塞米松(DEX)组、RU-38486组、逍遥散组、RU-38486联合逍遥散组。除模型组外,其余各组大鼠每天均在慢性应激刺激前2h腹腔注射DEX(0.1mg/kg),然后各组大鼠均给予慢性应激刺激,共21d。第21天,各组大鼠麻醉后,腹腔主动脉取血2mL,分离血清,采用酶联免疫吸附法(ELISA)测定大鼠血清皮质酮水平。结果:模型组大鼠血清皮质酮浓度显著高于其余各组(P<0.05),RU-38486组大鼠血清皮质酮浓度显著低于其余各组(P<0.01)。结论:RU-38486可能通过作用于海马神经元细胞的GR受体而抑制HPA轴的负反馈功能;中药复方逍遥散也可抑制HPA轴的负反馈功能,但其作用机制尚需进一步研究。
【Abstract】 Objective:To investigate the effect of Xiaoyaosan and RU-38486 (GR receptor antagonist )on the HPA axis negative feedback regulation function of chronic stress model rats. Methods:50 Wistar male rats were randomly divided into 5 groups:model group,dexamethasone (DEX)group,RU-38486 group,Xiaoyaosan group,Xiaoyaosan combined with RU-38486 group. Except model group,the rats in the other groups were injected with dexamethasone,2 h later were given chronic stress stimulation once a day for successive 21 d. Then blood samples were obtained from abdominal aorta of rats after anesthesia,serum corticosterone levels were measured by ELISA. Results:Serum corticosterone levels of rats in model group were significantly higher than those in the other groups (P<0.05);meanwhile serum corticosterone levels of rats in RU-38386 group were significantly lower than those in the other groups(P<0.01). There were no significant difference among DEX group,Xiaoyaosan group,Xiaoyaosan combined with RU-38486 group in serum corticosterone levels. Conclusion:RU-38486 can inhibit the function of HPA axis negative feedback regulation through acting upon GR receptor of hippocampal neuron;Xiaoyaosan has inhibitory effect on HPA axis,but further study is needed to find its mechanism.
