【摘要】 目的探讨细胞外基质蛋白1(ECM1)在结直肠癌组织中的表达及诊断意义。方法应用免疫组织化学检测结直肠癌手术切除的正常结直肠组织(n=20)、淋巴结未转移的结直肠癌(n=35)、淋巴结转移的结直肠癌(n=29)中ECM1蛋白的表达。Western blot检测ECM1亚型。受试者工作特征(ROC)曲线评估ECM1诊断价值。结果ECM1的表达在正常结直肠组织中阳性率为10%(2/20),在结直肠癌组织中阳性率为59.4%(38/64);其中结直肠癌淋巴结未转移者为37.1%(13/35),淋巴结转移者为86.2%(25/29)。ECM1在结直肠癌组织表达增高,在转移性结直肠癌中的表达高于非转移性结直肠癌(P<0.01)。结直肠癌细胞中高表达的ECM1蛋白主要是ECM1a亚型。结论ECM1蛋白在结直肠癌组织中过表达,与结直肠癌的淋巴转移相关。ECM1蛋白对结直肠癌尤其是转移性结直肠癌有较高的诊断价值。更多还原

【Abstract】 Objective To investigate the expression and significance of ECM1 protein in colorectal cancer tissue.Methods Immunohistochemical method was used to detect the expression of ECM1 in tissues from normal controls(n=20),colorectal cancer without lymphatic metastasis(n=35) and colorectal cancer with lymphatic metastasis(n=29).Western blot was used to identify the subtypes of ECM1.Results The positive rate of ECM1 was 10%(2/20) in normal colorectal tissues,59.4%(38/64) in colorectal cancer tissues,37.1%(13/35) in colorectal cancer tissues without lymph node metastasis and 86.2%(25/29) in those with lymph node metastasis.The ECM1 expression was higher in colorectal cancer tissues than in normal tissues and the expression in metastatic colorectal cancer was higher than that in primary colorectal cancer tissues(P<0.01).The highly expressed ECM1 protein was ECM1a subtype,with a relative molecular mass of 68 000.Conclusions ECM1 is over-expressed in colorectal cancer tissues,which is associated with the metastasis of colorectal cancer.ECM1 is a good biochemical marker in the diagnosis of colorectal cancer especially for the metastasis of colorectal cancer.更多还原
