胍丁胺对实验性变态反应性脑脊髓炎中枢神经系统内GFAP表达的影响

【摘要】 目的观察胍丁胺对实验性变态反应性脑脊髓炎(EAE)中枢神经系统内胶质纤维酸性蛋白(GFAP)表达的影响。方法用豚鼠全脊髓匀浆免疫Wistar大鼠建立EAE动物模型,于免疫当日开始分别予25、50、100mg/kg胍丁胺,腹腔注射,每天1次,连续14d。比较各组的行为学的变化,应用免疫组化法观察脑和脊髓组织中的GFAP的表达。结果与模型组比较,胍丁胺给药14 d EAE大鼠的发病率低,临床症状改善;EAE大鼠CNS内的炎性脱髓鞘病灶内和周围GFAP表达明显升高(<0.01)。结论胍丁胺对EAE具有保护作用。其机制可能与星形胶质细胞增殖活化有关。

【Abstract】 Objective To observe the effects of agmatine(AGM) on the expression of GFAP of the central nervous sys-tem(CNS) in experimental allergic encephalomyelitis(EAE).Methods EAE was induced in Wistar rats by immuniza-tion of spinal cords homogenate of guinea pigs(GPSCH) and complete Freund’s adjuvant(CFA).EAE rats gived with ag-matine was intraperitoneally injected with agmtine at dosage of 25、50、100 mg/ kg,respectively,once daily for success-ive fourteen days after immunization.The ethological change was observed and the number of GFAP in central nervous system was examined with the method of immunohistochemistry.Results Compared with EAE model rats,agmatine could decrease the incidence of EAE and alleviate its clinical manifestation.The number of GFAP in and around the in-flamatory demylinatve lession of CNS in EAE increased obviously(<0.01).Conclusion It is indicates that agmatine may have protective effect on EAE.Its mechanisms may be related to proliferating and activating astrocyte of central nervous system in EAE.
