活血药对血管紧张素Ⅱ致心肌肥大细胞内钙浓度变化的影响
Influence of drugs for promoting blood circulation on Ca2+ in myocardial cells and myocardial fibroblast induced by angiotensin Ⅱ
【摘要】 目的研究活血药丹参素和川芎嗪对血管紧张素Ⅱ诱导的乳鼠心肌细胞内和心肌成纤维细胞内钙离子浓度的影响,探讨其抑制心肌肥大的作用机理。方法应用Fura-3荧光染料,用流式细胞仪检测乳鼠心肌细胞和心肌成纤维细胞内钙离子浓度。结果AngⅡ可明显增加心肌细胞和心肌成纤维细胞内钙离子浓度,并随着时间的延长,于第5天其浓度达到高峰,然后下降,洛沙坦(Losartan)明显抑制其心肌和心肌成纤维内钙离子浓度的增加;研究显示丹参素和川芎嗪不仅明显抑制血管紧张素Ⅱ诱导心肌细胞内钙离子浓度的升高,同时也明显抑制血管紧张素Ⅱ诱导心肌成纤维细胞内钙离子浓度的升高。结论丹参素和川芎嗪不仅明显抑制血管紧张素Ⅱ诱导心肌细胞内钙离子浓度的升高,同时也明显抑制血管紧张素Ⅱ诱导心肌成纤维细胞内钙离子浓度的升高,表明丹参素和川芎嗪可能具有钙拮抗剂的作用,抑制血管紧张素Ⅱ所介导引起细胞内钙离子升高,从而抑制心肌细胞肥大和心肌成纤维细胞增殖。
【Abstract】 Objective To study the effective mechanism of Danshensu and Ligustrazine,the effective fractions in Chinese herbal medicines with drugs for promoting blood circulation,on Ca2+ in neonatal mice’s myocardial cells and myocardial fibroblast induced by angiotensinⅡ.Methods Ca2+ in myocardial cells and myocardial fibroblast has been detected by coolter EPICS XL with Fura-3.Results Not only can the Danshensu and Ligustrazine inhibit the increase of Ca2+ concentration of myocardial cells significandy, but also the proliferation of myocardial fibroblasts.The mechanisms may be Danshensu and Ligustrazine as Ca2+ antagonists,which reduce the increase of Ca2+ concentration induced by AngⅡ.Conclusion It showed that the drugs for promoting blood circulation decrease the high Ca2+ concentration induced by AngⅡ,so that they can inhibit hypertrophy of cardiac myocytes and hyperplasia of cardiac fibroblasts and prevent LVH.
