RP-HPLC法同时测定虎杖中虎杖苷、白藜芦醇、大黄素和大黄素甲醚的含量

【摘要】 目的建立同时测定虎杖中虎杖苷、白藜芦醇、大黄素和大黄素甲醚含量的分析方法。方法采用反相高效液相色谱法,以Diamonsil C18(4.6mm×250mm,5μm)为色谱柱,乙腈和0.1%磷酸为流动相梯度洗脱,对10个产地虎杖样品中虎杖苷、白藜芦醇、大黄素和大黄素甲醚的含量同时进行了测定。检测波长305 nm,流速1,0mL/min,柱温30℃。结果虎杖苷、白藜芦醇、大黄素和大黄素甲醚分别在8.39~83.85 mg/L、3.36~33.6 mg/L、3.09~30.9 mg/L和2.1 5~21.5 mg/L浓度范围内符合线性关系,其平均回收率分别为100.8%[相对标准差(relative standard deviation,RSD)1.3%]、99.3%(RSD 1.6%)、98.1%(RSD 1.5%)和92.2%(RSD 2.7%)。不同产地虎杖中的虎杖苷、白藜芦醇、大黄素和大黄素甲醚含量有较大的差别。结论本方法准确、简便、重现性好,可用于虎杖中虎杖苷、白藜芦醇、大黄素和大黄素甲醚含量的同时测定。

【Abstract】 Objective To establish reversed phase-high performance liquid chromatography (RP-HPLC)method for simultaneous determination of piceid,resveratrol,emodin and physcion in root of Polygonum cuspidatum.Methods The separation was performed in a Diamonsil C18 column with a gradient elution of acetonitrile and 0.1% H3PO4.The detection wave length was at 305 nm and the flow rate was 1.0 mL/min,the column temperature was 30℃.Results For piceid, resveratrol,emodin and physcion,the calibration curve was linear in the range of 8.39~83.85 μg/mL,3.36~33.6 μg/mL,3.09~30.9 μg/mL and 2.15~21.5 μg/mL,respectively.The linearity of this method was good.The average recovery was 100.8%(relative standard deviation,RSD=1.3%),99.3%(RSD=1.6%),98.1%(RSD=1.5%),92.2%(RSD=2.7%), respectively.In different producing area of Polygonum cuspidatum,the content of piceid, resveratrol,emodin and physcion were different.Conclusion The method is accurate,simple and reliable and can be used for quality control of Polygonum cuspidatum crude drugs.
