牛结核杆菌病PCR检测方法的建立及初步应用

【摘要】 根据PCR技术原理,建立了牛结核病PCR诊断方法,并测定出该方法的敏感性为1.025pg,菌细胞的灵敏度为200个菌左右。特异性试验表明,除牛结核分枝杆菌的特异扩增带(317bp)外,其他的细菌均未出现特异性扩增带。采用PCR检测方法,通过对94份奶牛奶样的检测,得到牛结核分枝杆菌的阳性率为11.7%(11/94),PCR和胶体金方法检出率有明显的差异。PCR方法敏感性强,特异性高,是一种提高牛结核病临床诊断的好方法。

【Abstract】 A PCR detection of Bovine tuberculosis was developed based on PCR principle with good sensitivity. The PCR reaction can be used to detect 1.025 pg Mycobacterium bovis DNA and 200 cells. The result of PCR specificity shows that the specific band (317 bp) exists only in Mycobacterium bovis but non in other bacteria. There were 94 cases diagnosed with the PCR detection and indicating that the positive infectious rate is 11.7% (11/94), which is significantly different to the colloidal gold test. With high sensitivity and specificity, the PCR is a good method for clinical diagnosis of bovine tuberculosis infected by Mycobacterium bovis.
