培养时间对尖孢镰刀菌产毒影响的初步观察
Preliminary observation on the effect of incubation time on the toxin production of Fusarium oxysporum
【摘要】 目的探索实验室培养尖孢镰刀菌产T-2毒素最佳条件。方法实验室用尖孢镰刀菌接种于普通琼脂培养基进行培养,8 d后分离、纯化,将第二代尖孢镰刀菌孢子在无菌条件下接种于灭菌后的市售玉米培养基(pH值7.0~7.2),培养温度为15℃,分别培养2周、4周和6周,采用高效液相色谱法对其培养物中的T-2毒素含量进行检测。结果实验室培养时间的不同,对T-2毒素的产量有明显影响,3种不同时间的培养物,其T-2毒素含量分别为6周≥4周≥2周。结论尖孢镰刀菌实验室培养时间的不同,对T-2毒素的产量有明显影响。
【Abstract】 Objective The aim of the study is to explore the optimum condition to cultivate the Fusarium oxysporum and produce T-2 fungal toxin.Methods The fungi were isolated and purified after the cultivation of 8 days,second filial generation sporus were inoculated and cultivated on sterile corn(pH 7.0~7.2)with different time(2w,4w,6w),the temperature was 15 ℃.Results The contents of T-2 fungal toxin were detected with high performance liquid chromatogram(HPLC).The result shows that the cultivated time is the important factor and the content of T-2 fungal toxin is different,6w≥4w≥2w.Conclusion The cultivated time is the important factor of the content of T-2 fungal toxin.
