葛根素对失血性休克凝血与纤溶功能的影响及其机制
Influence of Puerarin on the Blood Coagulation and Fibrinolyic Function of Rabbits with Hemorrhagic Shock
【摘要】 目的:观察葛根素对失血性休克家兔凝血与纤溶功能的影响及其机制。方法:将32只家兔随机分为假手术组、休克组、生理盐水组及葛根素组,每组8只。颈总动脉快速放血建立失血性休克模型。检测休克后0.5h、2h、4h的凝血酶原时间(PT)、活化部分凝血酶原时间(APTT)、血浆纤维蛋白原(FIB)与D-二聚体(DD)含量、红细胞超氧化物歧化酶(SOD)活性及血浆丙二醛(MDA)含量,观察葛根素对失血性休克凝血及纤溶功能的调节作用。结果:与假手术组比较,休克组PT、APTT时间缩短;FIB含量先升高后逐渐降低,D-二聚体含量逐渐增加;SOD的活性降低,MDA的含量增高;差异具有显著性(P<0.01)。休克后生理盐水复苏,PT、APTT 2h、4h略有延长(P<0.01);FIB无明显改变,D-二聚体含量略有下降(P<0.01或P<0.05);血浆MDA水平无明显影响,红细胞SOD活性在复苏4h后降低(p<0.05)。与休克组相比,用葛根索联合生理盐水治疗后PT、APTT时间延长,FIB含量和SOD活性均显著增高,D-二聚体含量与MDA含量显著降低(P<0.01),且作用效果强于单纯生理盐水组(P<0.01或P<0.05)。结论:葛根素能明显改善失血性休克时的凝血与纤溶功能紊乱,机制可能与抗氧自由基、改善内皮细胞功能有关。
【Abstract】 Objective:Investigate the effect of puerurin on the blood coagulation and fibrinolyic function of rabbits with hemorrhagic shock and discuss the mechanism. Methods: The model of hemorrhagic shock was established. 32 rabbits were randomly divided into four groups:sham operation group, shock group, normal saline group (NS group), puerarin group, and each group consisted of 8 rabbits. 0.5 hour, 2 hours and 4 hours after shock rabbits recepting puerarin,PT、APTT、FIB、DD、MDA、SOD activity in blood of rabbits were detected respectively. Results: compared with sham operation group, the results of PT, APTT in shock group reduced. With the development of shock, fibrinogen increased at 0.5 hour after shock and markedly redueed from 2nd hour (P<0.01). serum MDA and DD inereased (P<0.01), hut SOD decreased (P<0.01). After injection of NS, PT、APTT prolonged at 2、4 hours (P<0.01). Serum MDA and DD decreased, but FIB and SOD has no significant change. After injection of puerarin, PT、APTT prolonged evidently. Puerarin could reduce serum MDA and DD, increase FIB and superoxide dismutase activity(P<0.01). And the effect is stronger than NS group(P<0.01或P<0.05). Conclusions: Puerarin can obviously improve the abnormity of blood coagulation in rabbits with hemorrhagic shock, which is related probably to its antioxidant effect and functin improvement of endothelial cells.
