不同中药提取物对小鼠乳腺上皮细胞β-酪蛋白表达的影响

【摘要】 为探讨中药对小鼠乳腺上皮细胞泌乳功能的影响,选取5种与泌乳相关的中药(王不留行、甲珠、通草、蒲公英、漏芦)研究其对体外培养的小鼠乳腺上皮细胞β-酪蛋白表达的影响。采用荧光定量RT-PCR及毛细管电泳技术分别对β-酪蛋白的mRNA和蛋白表达水平进行检测,并应用激光共聚焦显微镜技术检测泌乳信号转录因子Stat5的含量变化。结果表明,通草、蒲公英和漏芦能显著促进β-酪蛋白的表达,王不留行和甲珠能极显著促进小鼠乳腺上皮细胞β-酪蛋白的表达。提示王不留行、甲珠和通草可通过影响乳腺上皮细胞乳蛋白基因的表达而促进泌乳,其机理尚待进一步研究。

【Abstract】 Five Chinese herbs were selected to study the lactating function of mouse mammary gland epithelial cells including cowherb seed, pangolin scales, medulla tetrapanacis, radix rhapontici and dandelion which were used extensively. Effects of herbs on the expression of β-casein of mouse mammary gland epithelial cells cultured in vitro were detected, respectively. Real time quantitative PCR was used to detect the expression of β-casein mRNA level and capillary electrophoresis technique was used to analyze the protein expression. Stat5 was detected by laser scanning confocal influorescence microscopy. The results showed that medulla tetrapanacis, dandelion and radix rhapontici can significantly induce the expression of β -casein, while cowherb seed and pangolin scales both can significantly induce the expression of β-casein extremely. It implies that cowherb seed, pangolin scales and medulla tetrapanacis can raise lactating qualities through impacting the secretion of milk protins of mammary gland. However, the mechanism remains to be further investigated.
