【摘要】 目的伐地那非是临床治疗男性勃起功能障碍(ED)的一线药物,但疗效在不同人群中差异较大。本研究旨在探讨ED患者PDE5基因第2外显子Val 93 Ala基因多态性对伐地那非疗效的影响。方法对本院270例ED患者给予伐地那非口服1周,同时采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法对所有患者进行PDE5基因分型,观察不同基因型治疗前后性功能指标变化、疗效及不良反应。结果270例ED患者中AA型为201例,AG型为44例,GG型为25例,基因型分布符合H-W平衡,具有群体代表性。伐地那非治疗后性生活持续时间、性生活满意度和IIEF评分各项指标在3种基因型之间差异有统计学意义,G等位基因携带患者有效率显著低于A等位基因携带患者。结论伐地那非治疗ED的疗效和个体遗传因素有关,PDE5基因G等位基因携带者疗效较差,应引起临床重视。更多还原

【Abstract】 Objective Vardenafil,a common medicine to treat the ED patients,the effect is different among different population.In this study,to explore the effect of vardenafil on the ED patients assotiated with polymorphism of PDE5 gene.Methods 270 patients with ED were divided into 3 groups accoring to the PDE5 genotypes identifided by the polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) methods.The changes of sexual function indexes,clinical effect,and untoward reaction after treatment with vardenafil for one week were compared among the 3 groups.Results There were 201 case with AA genotype,44 with AG genotype and 25 with GG genotype in 270 patients,the distribution of genotype was in H-W equilibrium.There were significantly different among 3 groups in vita sexualis duration,satisfaction degree and IIEF score after treated with vardenafil.The carrier with G allel had the lower effective power than those with A allel.Conclusion The effect of vardenafil on the ED patients assotiated with polymorphism of PDE5 gene.The patients with G allel of PDE5 gene has the lower effective power,which shoud be thought highly in clinic.更多还原
