HPLC法测定黄连盐酸小檗碱含量
Determination of Berberine Hydrochloride from Different Species of Coptis in Different Extracts Processess by HPLC
【摘要】 以湖北省来凤县中药材基地生产的3种黄连:味连(Coptis chinensisFranch),雅连(Coptis deltoidea C.Y.Chenget Hsiao),云连(Coptis teeta Wall)和利川市中药材基地生产的味连(Coptis chinensis Franch)为试材,以甲醇和乙醇为提取液,在85℃水浴中回流提取1.5h,用HPLC法测定比较不同种和同一品种不同产地的黄连盐酸小檗碱含量的差异。结果表明:云连小檗碱含量最高,雅连其次,川连最低,利川中药材基地生产的黄连中小檗碱的含量比来凤县生产的要高。溶剂种类对黄连盐酸小檗碱的提取率有一定的影响,甲醇作为提取液时的提取率相对较高一些。
【Abstract】 To establish the extraction method of berberine hydrochloride from coptis,To determine the content of berberine hydrochloride from different species of coptis and the same specie in different productive area of coptis.Develop a method of the highest extract rate,the extraction is the most efficient,and find a highest content specie of berberine hydrochloride of coptis to provide the quality basis for pharmaceutical preparation.Berberine hydrochloride from different species of coptis was determined by HPLC under the following condition:Column at C18(200mm×3.9mm 5μm),under low pH value,as mobile phrase using acetonitrile :0.1M KH2PO4(28∶72),with a flowrate at 1.0 mL/min,detection wave length at 346nm,column temperature at 25℃,aliquot injected 5μl.To observe the extract rate of berberine hydrochloride that effected by MeoH and Alcohol.Straction that subject spend this method backset current draw first and a half hours in heating in water bath at 80℃.: The content of berberine hydrochloride in different species was different.Among those Coptis teeta Wall was the highest,the following was Coptis deltoidea C.Y.Cheng et Hsiao and Coptis Chinensis Franch was the lowest.In extract rate,MeoH was higher than alcohol.HPLC is rapid,reliable,simple,sensitive and highly stable.The extract method is good for measuring the content of berberine hydrochloride in coptis.This method is worth popularinging.MenstruumVariety has great effect on the extract rate of berberine hydrochloride from Coptis.
