【摘要】 目的:研究化痰散结中药甲肿消对甲状腺细胞凋亡及Bax、Bcl-2表达的影响,探讨其治疗甲状腺肿大的作用机制。方法:甲肿消大鼠含药血清培养甲状腺FRTL细胞株,采用流式细胞仪检测细胞凋亡指数(AI%);实时荧光定量PCR法检测Bax、Bcl-2的mRNA表达,Western-Blot检测其蛋白表达。结果:甲肿消可以显著升高细胞的凋亡指数;大剂量和中剂量组Bax的mRNA和蛋白表达明显升高,具有统计学意义,Bax/Bcl-2的mRNA比值有升高趋势,中剂量组差异显著(P<0.05)。结论:化痰散结中药能促进甲状腺细胞凋亡,提示甲肿消治疗甲状腺肿大可能是通过促进细胞凋亡而实现,而Bax可能是其主要作用位点。更多还原

【Abstract】 Objective:To investigate the effect of removing phlegm and resovling mass Herbs Jiazhongxiao on apoptosis and expressions of bax、bcl-2 in FRTL cells. Methods: FRTL cells was cultured by Serum of Chinese Herb Jiazhongxiao. The Apoptotic Index(AI%)was tested by Fluorescence Microscope. The mRNA and protein expressions of Bax and Bcl-2 were detected with real-time quantitative RT-PCR and Western Blot. Results:The AI% increased remarkably because of Chinese Herb Jiazhongxiao. The mRNA and protein expressions of Bax increased distinctly in the DJ group (P<0.01) and ZJ group (P<0.05). The ratio of Bax/Bcl-2 increased remarkably in the ZJ group(P<0.05). Conclusion:This study showed that Chinese Herbs Jiazhongxiao can increase the apoptosis of thyroid cells, which may be related to the mechanism of Jiazhongxiao’ therapeutical effect on thyromegaly, and Gene Bax may be the key point.更多还原
