臭椿花粉生活力的测定方法及对比实验

【摘要】 利用氯化三苯基四氮唑(TTC)法、碘-碘化钾法、蓝墨水染色法、无机酸法、醋酸洋红染色法及离体萌发法测定了臭椿的花粉生活力。结果表明:碘-碘化钾法、蓝墨水染色法和醋酸洋红染色法都不适合臭椿花粉生活力的检测;TTC法,离体萌发法和无机酸法能有效快速地测定花粉生活力,但是TTC法与其它两种方法测得的结果差异显著。该实验结果认为离体萌发法和无机酸法更适合臭椿花粉生活力的检测。同时还讨论了6种测定方法的应用原理及优缺点。

【Abstract】 The pollen viability of Ailanthus altissima(Mill.)Swingle was detected by methods of triphenyltetrazolium chloride(TTC),iodine-iodide kalium(I-KI),blue-ink coloration(BIC),mineral acid(MA),carmine acetate dyeing(CAD),germination in vitro(GIV).The results showed that the methods of I-KI,BIC and CAD were not able to detect the pollen viability of Ailanthus altissima(Mill.)Swingle.The method of TTC,MA and GIV may detect pollen viability effectively and quickly,however,pollen viability were shown significantly different between TTC and MA/GIA.These results suggested that MA and GIV were fit for detection of pollen viability of Ailanthus altissima(Mill.).Furthermore,the application principles and their advantages and disadvantages of the six methods were discussed.
