脱细胞真皮基质修复硬腭软组织缺损的实验研究
Assessment of Acellular Dermal Matrix for Soft Tissue Reconstruction in Hard Palate
【摘要】 目的:研究同种异体脱细胞真皮基质(acellular dermal matrix,ADM)移植于硬腭黏骨膜与骨面之间,Ⅰ型胶原的变化情况,为其应用于腭裂软组织缺损的治疗提供实验依据。方法:取12周龄雌性Wistar大鼠60只,将大鼠硬腭黏骨膜从骨面剥离,将ADM基底膜面面向黏骨膜置入黏骨膜与骨面之间。分别于术后6、14、28、56 d随机处死动物15只,取硬腭部软组织标本行Ⅰ型胶原免疫组织化学染色观察,计数各时点标本中Ⅰ型胶原纤维束直径值,进行单因素方差分析。结果:ADM移植后Ⅰ型胶原纤维束直径值统计分析显示,Ⅰ型胶原纤维束逐渐进行改建,直径逐渐增加.移植后28 d达到正常真皮水平。结论:同种异体脱细胞真皮基质移植于硬腭黏骨膜和骨面之间.胶原改建良好,可以保护硬腭骨面免于坏死。
【Abstract】 Objective:To observe the variations of collagen typeⅠin acellular dermal matrix(ADM)after being transplanted beneath the mucoperiosteum of the hard palate.Methods:60 female 12-week-old Wistar rats were used for all experiments.Two lateral incisions were made along the gingival marginal of hard palate,and the mucoperiosteum was peeled off from the bone.ADM sheets were placed between mucoperiosteum and denuded bone,make the basement membrane surface towards mucoperiosteum.Every 15 rats were randomly sacrificed at 6th,14th,28th,56thday after the operation.Vaviations of collagen typeⅠwere detected by immunohistoehemistry.Results:The collagen fibers of ADM were grossness and arranged in horizontal direction 28 days later.Conclusion:ADM grafts survived well.The regeneration of collagen fibers were suit for the hard palate’s function,ADM could protect the denuded bone.
