HIF-1α、MMP-2的表达与膀胱移行细胞癌临床生物学行为的关系
Relationship Between the Expressions of HIF-1α and MMP-2 and the Biological Behavior in Human Bladder Transitional Cell Carcinoma
【摘要】 目的探讨缺氧诱导因子1α(HIF-1α)、基质金属蛋白酶-2(MMP-2)在膀胱移行细胞癌(BTCC)组织中的表达及其与临床生物学行为之间的关系。方法应用免疫组织化学检测BTCC组织中HIF-1α和MMP-2的蛋白表达,分析其表达与肿瘤部分临床病理特点的关系,并分析二者表达的相关性。结果正常膀胱组织中均未见HIF-1α和MMP-2蛋白表达,膀胱移行细胞癌组织中HIF-1α和MMP-2蛋白的阳性表达率分别为52.63%(30/57)和49.12%(29/57);HIF-1α阳性表达与肿瘤的临床分期及复发明显相关;MMP-2的阳性表达与肿瘤的病理分级、临床分期显著相关;HIF-1α的表达与MMP-2的表达密切相关。结论HIF-1α及MMP-2在BTCC的侵袭等生物学过程中具有重要的作用,HIF-1α和MMP-2也许可用于临床监控BTCC的进展。
【Abstract】 Objective To investigate the expressions of hypoxiainducibile factor 1α(HIF-1α)and matrix metalloproteinase 2(MMP-2) protein and their relationship with the biological behavior in bladder transitional cell carcinoma(BTCC). Methods HIF-1α and MMP-2 protein expression were determined using immunohistochemistry.The relationship between HIF-1α、MMP-2 expression and various clinicopathological parameters including pathological grade and clinical stage was analyzed and the correlation between HIF-1αexpression and MMP-2 expression was also analyzed. Results No marked expression of HIF-1α and MMP-2 was observed in the normal bladder tissues.The positive rate of HIF-1α and MMP-2 in BTCC was 52.63%(30 of 57) and 49.12%(29 of 57) respectively.HIF-1αprotein expression was significantly associated with patients,TNM stage and relapse(P<0.05).Significant differences existed in the expression of MMP-2 in carcinomas from different pothological grades and clinical stages(P<0.05).The expression of HIF-1α protein was significantly correlated with MMP-2 protein expression.Conclusion HIF-1α and MMP-2 may play a critical role in the invasion of bladder carcinoma and may be used to estimate the progression of bladder carcinoma.
