神经生长因子对机械性损伤大鼠脑皮质神经元的修复作用
Role of nerve growth factor in protection of centrifugation-induced rat cortical neuron injury
【摘要】 目的探讨神经生长因子(NGF)对机械性损伤大鼠脑皮质神经元的修复作用。方法分离培养大鼠脑皮质神经元及其微管相关蛋白-2(MAP-2)免疫荧光鉴定;通过台盼蓝拒染色动态观察NGF对神经元的活性的影响;采用机械性离心损伤皮质神经元,观察损伤后神经元的形态改变,并测量损伤前后不同时间段损伤组与损伤加NGF组乳酸脱氢酶(LDH)含量。结果培养4、5、6、8天NGF组与对照组进行比较,细胞存活率均有显著差异,P<0.05;损伤组与对照组LDH值比较差异有显著性(P<0.01);损伤组加NGF组与损伤组LDH值比较有显著差异(P<0.05,P<0.01)。结论NGF对皮质神经元有促生长作用;NGF对机械性大鼠脑皮质神经元的损伤具有修复作用。
【Abstract】 Objective To observe the role of nerve growth factor(NGF) in protection of centrifugation-induced rat cortical neuron injury.Methods The survival rates of primary cultural cortical neurons of rat were tested and the effects of NGF on the survival rates of neuron cells were observed through trypanblue dye assay;By means of centrifugating culture plate with vital neuron in vitro,the shapes of axons were observed,and lactate dehydrogenase(LDH) in culture medium at different stages were measured.Results In NGF group,the survival rates of primary cultural cortical neurons showed difference with control one(P<0.05).In injury group,LDH showed difference with control one(P<0.01).In NGF group,LDH showed difference with injury one(P<0.05,P<0.01).Conclusion The results suggest that NGF may promote the growth of cortical neurons cultured in vitro,NGF plays a protective role in the recovery of rat cortical neurons from centrifugation-induced injury.
