阿司匹林对MCF-7细胞uPA表达的影响

【摘要】 目的观察阿司匹林对乳腺癌细胞MCF-7增殖的影响,探讨阿司匹林新的抗肿瘤作用机制。方法MTT法检测不同浓度的阿司匹林(2.5、5.0和10.0 mmol/L)作用于人乳腺癌细胞株MCF-7不同时间(24、48和72 h)的细胞增殖抑制率;应用Western blot检测阿司匹林对MCF-7乳腺癌细胞尿激酶型纤维蛋白溶酶原激活剂(urokinase type plasminogen activator,uPA)表达。结果阿司匹林各剂量组细胞增殖抑制率随药物浓度的增加、作用时间的延长而提高(P<0.01);MCF-7细胞uPA的蛋白表达量随药物浓度的增加、作用时间的延长而降低。结论阿司匹林有抑制人乳腺癌细胞MCF-7增殖的作用,并且呈时间依赖性和剂量依赖性;阿司匹林可以抑制MCF-7细胞的uPA表达,这可能是阿司匹林抗乳腺癌的分子机制之一。

【Abstract】 Objective To explore the molecular mechanism of anti-tumor effects of aspirin on breast cancer cell MCF-7.Methods The human breast cancer cell MCF-7 was treated with 2.5,5.0,and 10.0 mmol ·L-1 aspirin for 24,48,and 72 h respectively,the inhibitory effect was detected by MTT assay.Western blot was used to observe the expression of uPA in MCF-7 cells treated by aspirin.Results The inhibitory rate of the MCF-7 cell line treated with aspirin was reduced compared with controls,which was in time and dose-dependent manner(P<0.01).The expression of uPA decreased after treated with aspirin,which also had a time and dose dependence.Conclusion Aspirin has inhibitory effects on the growth of human breast cancer cell MCF-7 in a dose and time dependent manner,and inhibits the expression of uPA in breast cancer cells,which is possible one of the molecular mechanisms of aspirin in breast cancer.
