复方当归汤中阿魏酸在兔体内的药动学研究
Study on Pharmacokinetics of Ferulic Acid in Compound Angelica Scinensis Solution Taken in Rabbit
【摘要】 目的:研究复方当归汤中阿魏酸在兔体内的药物动力学规律。方法:采用兔按4ml·kg-1单次灌胃给予复方当归汤,于不同时间点取样,不测定血浆中阿魏酸浓度,并用3P87软件处理药物动力学参数。采用RP-HPLC测定血药浓度,以甲醇-水(0.3%冰醋酸)(40:60)为流动相,Zorbax SBC18(150 mm×4.6 mm,5μm),紫外检测波长323 nm。结果:兔口服复方当归汤的血浆药-时曲线表现为单峰二室模型,主要药物动力学参数如下:Ka(min-1)=0.1400,K21=1.5×10-2,K10=1.1×10-2,K12=3.4×10-2,t1/2α(min)=12.05,t1/2β(min)=224.45,t1/2γ(min)=5.11,Cmax(ng·ml-1)=73.20,AUC(ng·min-1·ml)= 1.082×104,CL/F(s)(L·min-1)=3.700×10-4,V/F(c)(ng·ml-1)=3.2×10-2。结论:复方当归汤的体内有效成分阿魏酸吸收分布呈二室模型。
【Abstract】 Objective:To study the pharmacokinetic parameter of ferulic acid of Compound Angelica Sinensis solution in rabbits. Method:Compound Angelica Sinensis Solution was ig by 4 ml·kg-1 in tabbits.The concentrations of ferulic acid in plasma at different time points were detected.And the pharmacokinetic porameters were obtocined by 3P87 software package.Zorbax SBC18(150nn×4.6 mm,5μm) was used to detect the concerntration and methanol-water(containing 0.3% acetic acided)(40:60) was the mobile phase and UV detetion was at 323 nm.Result:The main pharmacokinetic parameters of Ferulic acid were as follows:Ka(min-1)=0.1400, K21=1.5×10-2,K10=1.1×10-2,K12=3.4×10-2,t1/2α(min)=12.05,t1/2β(min)=224.45,t1/2γ(min)=5.11,Cmax(ng·ml-1) =73.20,AUC(ng·min-1·ml)=1.082×104,CL/F(s)(L·min-1)=3.700×10-4,V/F(c)(ng·ml-1)=3.2×10-2.Conclu- sion:The absorption and the metabolism of FA in Compound Angelica Sinensis solution is fitted with two-compartment model.
