【摘要】 目的:从调节局部免疫、影响黑素细胞黑素合成和迁移3个方面研究外用中药对大鼠皮肤的作用。方法:用中药乙醇提取液外涂大鼠背部皮肤,3周后活检取材,用半定量RT-PCR检测酪氨酸酶(tyrosinase,TYR)、干细胞生长因子(stem cell factor,SCF)、干细胞生长因子受体(stem cell factor receptor,c-KIT)、白介素-2(interleukine-2,IL-2)和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)mRNA的含量。结果:中药3、5、6、7外用后,大鼠皮肤中SCF mRNA表达显著增加,分别为对照组的2.59倍、5.82倍、2.60倍和3.74倍(P<0.01);中药1、2、4使IL-2 mRNA表达水平降至对照组的42.9%、50.5%和68%(P<0.01或P<0.05);而中药1、2、3、4、7外用后,大鼠皮肤中TNF-αmRNA表达水平也降低,分别为对照组的25.7%、22.8%、51.7%、53.4%和51.5%(P<0.01或P<0.05);中药5还明显上调TYR mRNA的表达为对照组的2.41倍(P<0.01)。结论:外用中药治疗白癜风有效的机制可能与调节皮肤局部微环境有关。更多还原

【Abstract】 Objective:In order to investigate the effects of several traditional Chinese medicines(TCM) on rat skin from topical immunomodulatory,melanogenesis and migration of melanocyte.Methods:The rat skin were treated by the ethanol extracts of TCM separately twice a day,and ethanol was used as negative control.Three weeks later,the skin samples were obtained and the mRNA expressions of tyrosinase(TYR),stem cell factor(SCF),stem cell factor receptor(c-KIT),interleukin-2(IL-2) and tumor necrosis factor-α(TNF-α) were detected by RT-PCR.Results:The expression levels of SCF mRNA at the sites applied with the TCM NO.3,NO.5,NO.6,NO.7 were statistically higher than the control group(P<0.01);and there was a statistical significant decrease in IL-2 expression after the treatment of TCM NO1,NO.2 and NO.4(P<0.01 or P<0.05);furthermore,the TCM NO.1,NO.2,NO.3,NO.4,NO.7 reduced mRNA transcripts for TNF-α to about 25.7%,22.8%,51.7%,53.4% and 51.5%of the negative control respectively(P<0.01 or P<0.05);and only the medicine NO.5 revealed an upregulation of mRNA transcripts for TYR(P<0.01).Conclusion:The results suggest that the improvement of cutaneous microenvironment after topical TCM application may be associated with repigmentation of vitiligo.更多还原
