银杏达莫注射液抑制大鼠静脉血栓形成及作用机制探讨
Effect of Ginkgo leaf extract/dipyridamole injection on venous thrombosis in rats and the possible mechanism
【摘要】 目的:研究银杏达莫注射液对大鼠静脉血栓形成的影响,并探讨其作用机制。方法:建立大鼠静脉血栓模型和血瘀模型,分别测定血栓重量、凝血时间参数、优球蛋白溶解时间(ELT)和血液流变学等指标。结果:在静脉血栓模型大鼠,银杏达莫注射液低、中、高剂量组(0.9,1.8,2.25 mL·kg-1)能明显降低静脉血栓重量(P<0.05或P<0.01),低、中剂量能明显缩短ELT,但低剂量显著延长活化部分凝血活酶时间、凝血酶原时间和凝血酶时间(P<0.05或P<0.01)。在血瘀模型大鼠,银杏达莫注射液高剂量组(2.25 mL·kg-1)大鼠的全血黏度、血浆黏度、红细胞压积均显著低于模型大鼠(P<0.05或P<0.01)。结论:在一定剂量下,银杏达莫注射液能够抑制静脉血栓的形成,其机制可能与改善血液流变学性质和增加纤溶作用等有关。
【Abstract】 Objective:To investigate the effect of Ginkgo leaf extract/dipyridamole injection on venous thrombosis and the possible mechanism.Methods:The venous thrombosis and hyper-viscosity were induced in rats.The effect of Ginkgo leaf extract/dipyridamole injection on thrombosis was evalua- ted by determining thrombus weight,clotting time,euglobulin lysis time(ELT)and hemorrheology.Re- suits:Ginkgo leaf extract/dipyridamole injection markedly decreased the weight of venous thrombus at 0.9,1.8,2.25 mL.kg-1(P<0.05 or 0.01),shortened ELT at 0.9 and 1.8 mL.kg-1,and prolonged the prothrombin time(PT),activated partial thromboplastin time(APTT)and thrombin time(TT)at 0.9 mL·kg-1(P<0.05 or 0.01).The parameters of hemorrheology were significantly improved by the injection at 2.25 mL·kg-1(P<0.05 or 0.01).Conclusion:Ginkgo leaf extrac/dipyridamole injection can inhibit venous thrombosis by improving hemorrheology and fibronolysis.
