K562细胞多重耐药机制的实验研究
Study on the mechanism of multidrug resistance in K562 cell
【摘要】 目的探讨K562细胞多重耐药性形成的分子机制。方法采用MTT法分别测定K562及阿霉素耐药的K562细胞对顺铂的耐药程度,采用RT-PCR检测细胞表面MDR、MRP、GST及MGMT分子表达水平,并确定其与耐药性的关系。结果顺铂对K562细胞的抑制率明显高于阿霉素耐药的K562细胞,同时阿霉素耐药的K562细胞的表面MDR、MRP、GST及MGMT的表达明显增高。结论MDR、MRP、GST及MGMT的高表达是K562细胞形成多重耐药性的重要机制之一。
【Abstract】 Objective To explore the mechanism of multidrug resistance in K562 cell.Methods MTT colorimetric analysis was used to measure the inhibitory effect of cisplatin(DDP) on K562 cell and K562/ADM cell and the resistance index.RT-PCR was used to evaluate the mRNA expression of MDR1、MRP、GST and GMST genes.Results The inhibition rate of cisplatin(DDP) on K562 cell is higher than that of K562/ADM cell.Simultaneously the mRNA expression of MDR1、MRP、GST and GMST in K562/ADM cell is higher that of K562 cell.Conclusion The higher mRNA expression of MDR1、MRP、GST and GMST genes is the main causes of multidrug resistance in K562/ADM.
