四肽化合物CMS030抗移植排斥作用的实验研究
Study on the suppressive effects of CMS030 tetramer on allograft rejection in mice
【摘要】 目的:CMS030是从脾脏提取的四肽化合物,本研究观察CMS030对小鼠皮肤和心肌移植排斥反应的影响。方法:通过体外T淋巴细胞转化实验和混合淋巴细胞反应(MLR)实验,观察CMS030在体外的免疫抑制作用,建立小鼠皮肤移植和心肌移植模型,观察CMS030对小鼠同种移植物存活时间的影响,观察CMS030对移植受体小鼠T淋巴细胞转化和脾细胞分泌IL-2的影响。结果:CMS030在体外能够抑制T淋巴细胞转化和MLR,在体内可以明显延长小鼠皮肤移植物和心肌移植物存活时间,抑制皮肤移植受体小鼠T淋巴细胞转化能力和脾细胞分泌IL-2水平。结论:CMS030通过抑制T淋巴细胞分泌IL-2,抑制T淋巴细胞功能,有效拮抗移植排斥反应。
【Abstract】 Objective:To investigate the effects of CMS030 on skin and cardiac allograft rejection in mice.Methods:The effects of CMS030 in vitro were studied by ConA induced T cell proliferation and mixed lymphocyte reaction(MLR). The tail skin grafts of BALB/c mice were placed in wound bed on the back of C57BL/6 mice, and the cardiac allografts of new-born C57BL/6 mice were transplanted into the posterior of BALB/c mice.The effects of CMS030 on the survival time of the grafts were observed. ConA-induced lymphocyte proliferation and the IL-2 of the mice were examined.Results:The inhibition effects of CMS030 on T cells were displayed in vitro and the survival time of grafts was significantly prolonged by CMS030 in vivo. ConA-induced lymphocyte proliferation and the IL-2 secretion of the mice were both reduced by CMS030.Conclusion:CMS030 can inhibit IL-2 secretion of T lymphocytes, and suppress allograft rejection efficiently.
