滋肾方对阿尔茨海默病模型大鼠学习记忆的影响
Effect of Zishen Prescription on Learning and Memory Ability of AD Rats
【摘要】 目的观察滋肾方对阿尔茨海默病(AD)模型大鼠学习记忆等行为学及海马组织胆碱酯酶(CHE)和单胺氧化酶(MAO)的影响,探讨滋肾方治疗AD的作用机制。方法持续注射D-半乳糖法致亚急性衰老大鼠模型,根据脑立体定位图谱,用鹅膏蕈氨酸(IA)化学损毁大鼠双侧Meynert基底核。以"Y"型电迷宫进行学习记忆观察,用分光光度法测定大鼠海马CHE和MAO的活性。结果与AD模型比较,滋肾方可以减少模型大鼠学习、记忆达标所需的电击次数[分别为(61.00±11.01)次,(43.70±11.02)次;(32.90±10.02)次,(23.40±3.13)次],差别有统计学意义(均P<0.01);提高模型大鼠海马组织中CHE活性[(4.23±0.45)U/mgpro,(5.33±0.57)U/mgpro],降低MAO活性[(12.81±1.56)U/h·mgpro,(8.90±0.93)U/h·mgpro]差别有统计学意义(均P<0.01)。结论滋肾方可以提高AD模型大鼠学习记忆能力,其作用机制可能与大鼠海马组织中CHE和MAO活性改变有关。
【Abstract】 Objective To observe the effect of Zishen prescription on the learning and memory abilities and activities of CHE and MAO in nerve tissues of the AD rats,and discuss the anti-AD effects of Zishen prescription.Methods The subacute aging rat models were made by injecting D-galactose;and then both sides of NBM were damaged with IA according to the brain stereotaxic atlas of rats.The learning and memory abilities were determined by ″Y″ maze;and the activities of CHE and MAO in the hippocampus of rats were tested by spectrophotometer.Results Compared with the results of AD control group,the treatment of Zishen prescription significantly decreased the number of electric stimuli(61.00±11.01 vs 43.70±11.02,P<0.01;32.90±10.02 vs 23.40±3.13,P<0.01),and the activity of MAO in the hippocampuses(5.33±0.57 U/mgpro vs 4.23±0.45 U/mgpro,P<0.01),but increased the activity of CHE(8.90±0.93 U/h·mgpro vs 12.81±1.56 U/h·mgpro,P<0.01).Conclusion Zishen prescription can improve the learning and memory abilities of AD rats,and the mechanism may be related to the decreased activity of MAO and increased activity of CHE in the hippocampuses of AD rats.
