骨髓基质细胞移植对大鼠脊髓损伤后胶质纤维酸性蛋白和神经丝蛋白表达的影响

【摘要】 目的:研究骨髓基质细胞(BMSCs)移植对大鼠脊髓损伤(SCI)后胶质纤维酸性蛋白(GFAP)和神经丝蛋白(NF)表达的影响,探讨BMSCs促进SCI修复的机制。方法:Wistar大鼠60只,随机分为移植组和对照组,伤后1,3,7,14,28d,每组每个时相点6只动物,用免疫组织化学染色方法观察GFAP和NF的表达。免疫组化结果用计算机辅助的数字图像分析仪进行定量分析。结果:BMSCs移植后可在损伤脊髓内生存、整合、迁移;BMSCs移植后在不同时间点均可促进损伤区周围脊髓GFAP和NF的表达。结论:BMSCs移植可通过促进损伤区周围脊髓GFAP和NF的表达等机制促进脊髓损伤修复。

【Abstract】 Objective: To explore the influence of bone marrow stromal cell(BMSCs) transplantation on expression of GFAP and NF in spinal cord injury of rat. Method: Sixty rats were divided into transplantation group and control group. The expression of GFAP and NF were evaluated at 5 time points by means of immunohistological method after transplantation or operation. Result: BMSCs could survive, integrate and immigrate in spinal cord after transplantation. The expression levels of GFAP and NF in transplantation group were significantly higher than those in control group on the 1st,3rd,7th,14th and 28th d. Conclusion: BMSCs can promote the expression of GFAP and NF, which can promote recovery of spinal cord injury.
