HSPA12A mRNA在难治性癫痫患者脑组织中高表达
Up-regulated expression of HSPA12A mRNA in the brains of patients with intractable epilepsy
【摘要】 目的利用cDNA芯片和荧光定量PCR(Fluorescence Quantitive Polymerase Chain Reaction,FQ- PCR)技术检测难治性癫痫患者脑组织中HSPA12A(heat shock 70kDa protein 12A,HSPA12A)mRNA的表达,并探讨其在难治性癫痫发病机制中的作用。方法收集36例难治性癫痫患者术后脑组织和8例正常对照组,用含有4096条人类靶基因的cDNA芯片进行扫描,筛选出候选基因后,用FQ-PCR,以3-磷酸甘油醛脱氢酶(GAPDH)为内参对芯片扫描结果进行验证。结果发现候选基因HSPA12A mRNA在难治性癫痫患者脑组织中表达上调,FQ- PCR与cDNA芯片结果一致。结论虽然HSPA12A具体生物功能尚不明确,但我们认为HSPA12A可能作为重要的调节因子参与了神经元凋亡等分子事件。HSPA12A mRNA表达上调可能在难治性癫痫发病机制中起一定作用。
【Abstract】 Objective To study the expression of HSPA12A(heat shock 70kDa protein 12A,HSPA12A) mRNA in the brains of patients with intractable epilepsy(IE)by using cDNA chip scanning and fluorescence quan- titive polymerase chain reaction(FQ-PCR),and discuss its biological function in the pathogenesis.Methods The surgically removed brain tissues of patients with IE(n=36)and normal controls(n=8)were collected,and scanned to detect differentially expressed genes by using cDNA chip with 4096 human target genes.Then the can- didate gene was selected,and FQ-PCR with gylceraldehyde-3-phosphate dehydrogenase(GAPDH)as internal con- trol was used to confirm the selected gene.Results The candidate gene of HSPA12A mRNA was higher ex- pressed in the brains of patients with IE,and the results of FQ-PCR were coincided well with the cDNA chip re- sults.Conclusion Further study should be made to clarify the precise function of HSPA12A.HSPA12A may he an important modulator taking part in the molecule event of neuron apoptosis.The results indicate that the up-reg- ulated expression of HSPA12A mRNA may play an important role in the epileptogenic mechanisms of IE.
