脱水-脑萎缩-可逆性的实验研究

【摘要】 目的通过动物实验静脉注射脱水剂甘露醇后观察脑组织细胞在电子显微镜下的变化,探讨其脑萎缩表现及可逆性。方法将实验杂交家兔44只分成健康组1组4只,对照组10组各4只;健康组兔不用脱水剂,对照组兔8组在不同时间段内静脉注入脱水剂后、对照组兔2组先脱水、补液后予以处死取兔脑组织标本做电子显微镜检,看细胞学变化。结果脱水剂20%甘露醇在大剂量使用早期就可出现对照组兔脑神经元细胞的胞质水肿、线粒体浊肿、粗面内质网及核糖体减少表现;脱水12d后可见部分神经元细胞变形,胞质内细胞器大部破坏、消失等改变。脱水14d及16d组兔神经元细胞变形坏死,核缩小,胞质内细胞器破坏、消失,有2只见神经元细胞有核皱缩改变。脱水8d后补充液体3d、脱水10d补充液体6d后发现仅有少部分线粒体有肿胀,神经元细胞核膜完整。结论通过实验电子显微镜下观察表明大剂量脱水剂的使用可使脑组织细胞出现损害现象;在一定时间内脱水后再补液脑组织细胞可逐步恢复正常。

【Abstract】 Objective To evaluate the effect of cell ultrastructure and learn the possible of brain atrophy and learn by some means brain atrophy can be recovered with rabbit brain by mannitol injection in vein with an animal experimental study. Methods 44 cases of local,healthy,crossbred rabbits,they were randomly divided into groups: one group called healthy group,including four animals without mannitol,the other groups called control group,including 10 groups,each with four animals. Every animal was injected mannitol from time to time,the last two groups were given fluid therapy intravenous after mannitol. All animals were died to bring brain tissues inspected with electron microscope to learn cell ultrastructure. Results In the early phase,animals were injected 20% mannitol,their sytoplasm edema,mitochondrion swell、rough endoplasmic reticulum and ribosome decrease in the control groups never cell,the 12th day partial never cell deform and large partial cell organelle broken and diminish,the 14th and 16th never cell deformed and necrosis,cell nuclear atrophy,cell organelle damage and diminish,pyknosis can be seen in two animals. The last two groups,one group was given fluid therapy intravenous 3 days after mannitol 8 days,the other group was given fluid therapy intravenous 6 days after mannitol 10 days can only seen small partial of mitochondrion swell. Conclusion The brain tissues cell can be injured by electron microscope with injection mannitol,and can be recovered given fluid therapy intravenous after mannitol in the special time.
