姜黄素对肺纤维化大鼠结缔组织生长因子表达的影响

【摘要】 目的研究姜黄素对博来霉素诱导肺纤维化大鼠结缔组织生长因子表达的影响。方法SD雄性大鼠72只,分为对照组、模型组、姜黄素治疗组,每组24只。对照组大鼠气管内一次性滴注等体积的生理盐水,其余各组大鼠气管内一次性滴注盐酸博来霉素。造模后第1天姜黄素治疗组开始给药为200mg.kg-1.d-1,持续到处死动物的前1d。对照组、模型组分别灌服等体积的生理盐水。各组动物于7、14、28d随机处死8只,取肺组织进行HE、Mallory染色,同时进行免疫组织化学染色,结合图像分析来检测各组肺组织中结缔组织生长因子蛋白的表达水平。结果模型组大鼠结缔组织生长因子表达水平与对照组比较显著增高(P<0.01);姜黄素治疗组大鼠结缔组织生长因子表达水平与对照组和模型组比较差异有统计学意义(均为P<0.05)。结论姜黄素具有抗肺纤维化形成的作用,可能与其抑制结缔组织生长因子在肺组织中的表达有关。

【Abstract】 Objective To investigate the effects of curcumin on the expression of connective tissue growth factor(CTGF)in rat pulmonary fibrosis induced by Bleomycin.Methods Seventy-two male SD rats were randomized into three groups:sham-model control group(n=24),model group(n=24)and curcumin-treated group(n=24).Rats in the model group and curcumin group were given a single dose of intratracheal bleomycin,except for the sham-model group which used the same volume of normal saline as control.Oral curcumin(200 mg·kg-1·d-1)was administered daily in the treatment group by gavage starting from day 1 until the day before animals were killed,while the other groups received equal volume of normal saline in the same way.On days 7,14 and 28 days,every 8 rats randomly chosen from each group were sacrificed and removed of lungs to be sectioned and stained with HE,Mallory methods and immunohistochemical techniques.Expression of CTGF in the lung tissue was analyzed using a computerized image system.Results Compared with the sham-model group,significant over-expression of CTGF was shown in lung tissue of the model group(P<0.01).Expression of CTGF in the curcumin-treated group was significantly higher than that in the sham-model group,and lower than that in the model group(both P<0.05).Conclusion The effects of curcumin on rat pulmonary fibrosis may be correlated with inhibition of CTGF over-expression in the lung tissue.
