【摘要】 目的:观察阿司咪唑对大鼠离体心脏心电图的影响,了解其心脏毒性作用。方法:应用Langen-dorff离体大鼠心脏灌流装置,分别用3种不同浓度的阿司咪唑(1×10-7、2×10-6、5×10-6mol/L)灌注,观察离体大鼠心脏心电图的变化。结果:2×10-6mol/L和5×10-6mol/L的阿司咪唑可诱发多种心律失常,并随剂量增加诱导时间明显缩短,心律失常的严重程度明显加重。且主要引起缓慢性心律失常,使心率明显减慢,PR、QT、QRS间期显著延长。而且这些作用均随药物剂量增加和作用时间延长而增强。结论:阿司咪唑对大鼠离体心脏具有毒性作用,主要引起缓慢性心律失常。更多还原

【Abstract】 AIM: To observe effects of astemizole on electrocardiogram in isolated rat hearts to study its cardiotoxicity. METHODS:By using the Langendorff device for isolated rat hearts, astemizole of different concentrations (1×10-7,2×10-6,5×10-6 mol/L) were perfused into rat hearts. Then observe the change of electrocardiogram of isolated rat hearts. RESULTS:2×10-6 and 5×10-6 mol/L astemizole could induce polymorphic arrhythmia. Its delitescence was shortened evidently and the arrhythmia became too much severe with the increasing of the dose. It could also slow heart rate markedly, induce bradyarrhythmia mostly, delay PR, QT, QRS interval evidently. These actions also intensified with the increasing of dose and prolonging of action time. CONCLUSION: Astemizol has cardiotoxicity to isolated rat hearts. It can induce bradyarrhythmia mostly.更多还原
