杂色曲霉素对小鼠穹隆下器官TNFα和TGFβ2的mRNA表达的影响
Effects of sterigmatocystin on mRNA expressions of TNFα and TGFβ2 in subfornical organ of mice
【摘要】 目的观察杂色曲霉素(ST)损伤脑组织时有无肿瘤坏死因子(TNF)α和转化生长因子(TGF)β2的mRNA表达的改变。方法用BALB/c小鼠,ST3mg.kg-1灌胃,分别于灌胃后0.5,1,2,4,8和16h处死动物,用原位杂交方法观察穹隆下器官(SFO)细胞TNFα mRNA和TGFβ2 mRNA的表达。结果TNFα mRNA和TGFβ2 mRNA阳性细胞广泛分布在SFO。ST处理组从0.5h开始TNFα mRNA阳性表达细胞数缓慢上升,4h达到顶点,之后缓慢下降;TGFβ2 mRNA阳性表达细胞数从0.5h缓慢持续上升,16h达到顶点。ST组各时间点TNFα mRNA和TGFβ2 mRNA阳性细胞数均明显高于对照组。结论TNFα和TGFβ2在ST对SFO细胞损伤和修复中可能起着重要的作用。
【Abstract】 AIM To investigate whether the expressions of tumor necrosis factor(TNF)α mRNA and transforming growth factor(TGF)β2 mRNA were changed after the injury of brain tissue by sterigmatocystin(ST). METHODS BALB/c mice were treated with ST 3 mg·kg-1, ig. The expressions of TNFα and TGFβ2 mRNA in subfornical organ(SFO) cells were detected by in situ hybridization 0.5, 1, 2, 4, 8 and 16 h after administration. RESULTS The positive cells of TNFα and TGFβ2 mRNA were seen in SFO. The number of TNFα mRNA positive cells increased slowly after administration ST 0.5 h, reached its peak at 4 h and then began to decrease. The number of TGFβ2 mRNA positive cells increased slowly after administration ST 0.5 h, reached its peak at 16 h. The rates of positive TNFα mRNA and TGFβ2 mRNA cells of ST treated groups were significantly higher than those in control group. CONCLUSION TNFα and TGFβ2 might play a critical role in the injury and reparation of SFO cells by ST.
